To study the relationship between expression of keratin 19 and pathological factors of nasopharyngeal carcinomaObjective To study and analsis the relationship between expression of keratin 19 (CK19) and pathological factors of nasopharyngeal carcinoma (NPC)Methods All specimens of 62 of NPC,17 of lymph node metastasic NPC (LMNPC) and 16 of chronic rhinitis (CR) (serve as normal control) were pathological confirmed and the expression of CK19 was detected by immunohistochemistry S-P method, and the correlation between the expression of CK19 and clinic factors were analysized by SPSS software. The expression of CK19 in NPC cell lines with differential differentiation degree and metastasis potency were detected by western blot.Results 1) The result of immunohistochemistry showed that CK19 was locaed in endochylema and cell membrane. The positive ratio of CK19 in NPC, LMNPC and CR were 74.2%(46/62),88.2%(15/17) and 12.5%(2/16), respectively. The expression of CK19 in LMNPC were higher than in NPC, in NPC than in CR (P<0.05).2) There were relationship between the expression of CK19 and differentiation of NPC. The positive ratio of CK19 expression in moderate-differentiated NPC and poorly-differentated NPC were 46.2%(6/13) and 81.6%(40/49), respectively. And there were significant difference between them (P<0.05).3) There were relationship between the expression of CK19 and primarily tumor size of NPC. The positive ratio of CK19 expression in non-invaded NPC and invaded NPC were 20.0%(1/5) and 78.9%(45/57), respectively (P<0.05)4) There were relationship between the expression of CK19 and lymph node metastasis of NPC. The positive ratio of CK19 expression in non-metastasic NPC and lymph node metastasic NPC were 11.1%(1/9) and 84.9%(45/53), respectively (P< 0.05).5) There were relationship between the expression of CK19 and distant metastasis of NPC. And the positive ratio of CK19 expression in non-distant-metastasic NPC (MO stage) and distant-metastasic NPC (M1 stage) were 71.4%(40/56) and 100%(6/6), respectively (P<0.05)6) There were relationship between the expression of CK19 and clinicâ… +â…¡stage and III+IV stage. And the positive ratio of CK19 expression inâ… +â…¡stage and III+IV stage were 45.5%((5/11)) and 78.8%((41/52), respectively (P<0.05)7) There were not relationship among the expression of CK19 and gender and ages of NPC.8) Western blot result showed that the expression of CK19 in poorly-differentiated CNE2 cells were higher than in well-differentiated CNE1 cells; in high metastasic 5-8F cells higher than in low metastasic 6-10B cells (P< 0.05). there were relationship between the expression of CK19 and differentiation and metastasis of NPC.Conclusion1) The expression of CK19 in LMNPC were higher than in NPC, in NPC higher than in CR or normal mucous membrane of nasopharynx; The expression of CK19 were more higher in poorly-differentiation and high metastasic potentiality NPC cell lines than in good-differentiation and low metastasis potentiality NPC cell lines. And CK19 could serve as a biomarker of differentiaion and metastasis of NPC.2) There were correlation among the expression of CK19 and differentiation degree, primary tumor stage, lymph node meastasis and distant metastasis, clinic stage. And without relationship to ages and gender.
