1. Objective1.1 Explore the method of in vitro on-site determination about Plasmodium falciparum to current artemisinin-based combination antimalarial drugs; to provide dihydroartemisinin/piperaquine measurement board with medicinal stable, repeatable, valid for not less than 3 months.1.2 We monitor sensibility in Plasmodium falciparum endemic area of Yunnan Province, provide technical support for surveillaneing the sensitivity of artemisinin-based combination antimalarial drug in China (dihydroartemisinin /piperaquine) to Plasmodium falciparum,1.3 Find out dihydroartemisinin/piperaquine compatibility with (or without) synergies2. MethodsThe method in vitro microtest current used was standardized from the WHO in vitro drug sensitivity of detection. This method is based on in vitro culture method by Trager and Jensen and evolved in 1978 to further establish in vitro microtest for antimalarial drugs sensitivity by Rieckmann. It is based on the different concentration of drugs makes different impact on the parasite growth and development, drugs can completely inhibit Plasmodium falciparum growth and development by certain concentrations. It can evaluate the inhibitory action of anti-malarial drugs on the formation of schizont in vitro by microscopic examination.2. ResultsThe success rate of single-drug determination plates is 76.92%.75% in 2008 and 72.41%,75.86% in 2009. The success rate of compound-drugs determination plates is 76.92%(2008) and 75.86%(2009); the difference was not statistically significant. The inhibited parasite rate of each Unilateral and compound well was in turn increased with drug concentration increased. The compound drugs inhibite the parasite significantly higher than unilateral, compound drugs'half inhibitory concentration (IC50) was significantly lower than unilateral, the compound drugs significantly reduce the amount of drug than unilateral.4. Conclusions4.1 It is suggested that self-restraint dihydroartemisinin/ piperaquine drug-coated plates have stabilization, sensitiveness, good repeatability and user-friendly and can promote the use of the on-site.4.2 It is significantly reduced the drug dosage when combined with the two drugs and may delay the emergence of drug-resistant, so it is imperative combined.
