Objective: To detect the miRNA34b tumor suppressor gene in non-Hodgkin,s lymphoma of the promoter methylation status and to approach the relationship and significance between miRNA34b gene methylation and non-Hodgkin,s lymphoma..Methods: (1) To choose 70 samples of non-Hodgkin,s lymphoma and 20 samples of benign disease of Lymphoid tissues in first affiliated hospital department of oncology.(2)The use of matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis detected 70 cases of non-Hodgkin,s lymphoma and 20 cases of benign disease of Lymphoid tissues in the methylation status of miRNA34b gene . Analysis the difference in non-Hodgkin,s lymphoma with gender , age, and histological. All data were analyzed by using the SPSS 16.0 software.Results: 1.MiRNA34b gene methylation test results: (1)Amongst the 70 cases of non-Hodgkin,s lymphoma started, there are 52 males and 18 females.The male-to-famale ratio was 2.89:1. The media age of the patients was 65 years.(2) There was no relationship between the miRNA34b gene methylation level and the gender and age of the patients with non-Hodgkin,s lymphoma(P>0.05).(3)70 cases of non-Hodgkin,s lymphoma in the level of miRNA34b gene methylation was higher than benign disease of lymphoid tissues (P<0.05), the average rate of methylation (28.5%) higher than the control group the average rate of methylation (12.4%) (P <0.05). The average methylation rate of miRNA34b -16CpG1.2.3, CpG11.12.13, CpG14 were higer than 35%. (4) The average methylation rate of miRNA34b in four different histological subtypes has statistical significant (P<0.05). (5)The diffences in three type of B cell lymphoma has statistical significant (P<0.05).The average methylation rate of diffuse large B cell lymphoma was obvious higher than that of the three other B cell lymphoma.Conclusion: (1) The methylation of miRNA34b gene in non-Hodgkin,s lymphoma is a frequent event., and miRNA34b gene methylation have connection with the development of non-Hodgkin,s lymphoma.(2) The methylation level of miRNA34b maybe has important relationship with the malignant degree of the cancer in B cell lymphoma.
