Ohjective:To investigate the effect of GM6001 on corneal damagement and restoration after laser epithelial keratomileusis, meanwhile, to explore the mechanism of corneal healing after laser epithelial keratomileusis.Methods:(1)To make up the GM6001 eye drops (2)To build the LASEK models of rabbits (3)Twenty-seven New Zealand white rabbits were divided randomly into GM6001 group,FML group and NC group, Corneal haze grade were evaluated by slim light at 0.5,land 2 mouth postoperative intervals. Two rabbits were randomly harvested in each group at different postoperative intervals and then examined by optical and transmission electron microscope(TEM),confocal microscope and collagen typeⅢexpression by immunohistochemical analyses.Results:1. Slim light of corneas:Corneal haze grades at 0.5 and 1 month after surgery were significant differences among the groups(P<0.01),Comparing with the NC group,The Corneal haze grades in GM6001 group and FML group were apparently lower(P<0.01),but there was no significant difference between GM6001 and FML groups(P> 0.05).And there was no significant differences among the three groups at 2 month postoperatively as well(P>0.05).2. Pathological section of corneal tissue:The corneal epithelial cells of experimental groups were hyperplasiaed thick evidentiy at each postoperative interval. The corneal epithelial cells were increased to 6 or 7 layers, Comparing with the NC group, the corneal stroma cells were less concentrated at the ablation zone in GM6001 group and FMLgroup.3. collagen typeⅢexpression by immunohistochemical analyses:Negative expression of collagen typelll in normal cornea. At one month after surgery, collagen typelll was strong expressed in NC group. At one month after surgery,collagen typeⅢwas weakly expressed in GM6001 group and collagen typelll was positively expressed in FML group.4. transmission electron microscope(TEM):Comparing with the NC group and FML group, the numbers of fibroblast at the fore-stroma were less, the collagen fibers keeped well-arranged and the capacities of fibroblasts were small in corneal stroma in GM6001 group, the contents of cellular organ were few and cells were stable in GM6001 group.5. Confocal microscopy through focusing(CMTF):Contrast the GM6001 group with the NC group and FML group, the corneal stroma cells were aligned more regular. The numbers of keratocytes in GM6001 group were apparently lower than in NC group and in FMLgroup(P<0.01), and there was difference between NC group and FMLgroup(P<0.01).Conclusion:GM6001 has inhibitive effect on corneal haze formation after LASEK, which by suppressed the proliferation of keratocytes and decreased the synthesis of collagen typeⅢ.This efficacy was similar to FML. The mechanism of corneal haze formation was relevant to release MMPs after operation that caused damage of cornea, which inducing the proliferation of keratocytes and leading to collagen typeⅢarrayed disorderly.
