| Objective:Researchers all over the world have been studying on methods to repair articular cartilage defect for many years. There is a growing body of evidence that proves growth factors play an important role in the physiological processes such as proliferation, division and metabolism of chondrocytes by regulating signals. The cartilage inductive capacity of demineralized osteoarticular allograft transplantation in the repair of articular cartilage injuries has been supportted by a large number of experiments. In this research demineralized osteoarticular allograft transplantations in the patellar grooves of left knee joints of New Zealand white rabbits were finished seperatively. By means of the technique of proteomics we analyzed the emerging proteins existing in synovia after transplantation in order to find the specific proteins in the repairing process and investigate the mechanism and mode of cartilage reproduction.Methods:Demineralized osteoarticular allografts whose articular cartilage was protected were produced at first. In 20 New Zealand white rabbits, articular cartilage defects of 4mm diameter and 3mm depth were created in the patellar grooves of left knee joints. And then all the rabbits were divided into 2 groups randomly. Demineralized osteoarticular allografts were transplanted into the articular cartilage defects in groupâ… . As controls, all the defects in groupâ…¡were left alone. After transplantation, synovia samples of 2 groups were collected at once and in the 8th, 12nd , 16th and 24th week. Two-dimensional electrophoresis(2-DE)of the synovia samples was conducted, and then distinct protein spots were identified by means of the matrix-assisted laser desorption/ionization tandem time-of-flight mass spectrometry (MADLI-TOF-TOF-MS).Results:Rabbit models of articular cartilage defects were established successfully. The repairing progress of articular cartilage injuries was induced by demineralized osteoarticular allograft transplantation and thousands of emerging proteins were isolated through 2-DE of synovia samples. 23 significative differential protein spots were found in gel-analysis as compared with controls according to the experiment principle. The further MADLI-TOF-TOF-MS analysis revealed three peculiar kinds of proteins whose GenInfo identifier number were 187735841,163796533 and 736808 might be associated to the regeneration of articular cartilage injuries.Conclusion:Three specialized kinds of proteins which may play important roles in the articular cartilage reproduction were identified in the process of demineralized osteoarticular allograft transplantation. One kind of tape measure protein whose GenInfo identifier number was 187735841 may be contacted with the maintenance of cells function and activation of gene expression. Another kind of DNA/RNA helicase protein with the GenInfo identifier number 163796533 is a member of SNF2 family and may affect the progresses of gene duplication, repair and expression. The other kind of paraoxonase protein with the GenInfo identifier number 736808 has the antioxidant ability to prevent cells from oxidative injury efficiently. This finding may establish a foundation to the further research. |
