Experimental Study On The Effect Of Tianma Granules On Apoptosis And Related Gene Of Human Colorectal Cell Line Sw-480

Objective:to observe the effect of Tian Ma Granules on apoptosis of human colonic carcinoma cell line SW480, and to observe the gene expression of wpP53 and bcl-2.Methods:Model rats of transplantation tumor of human conlonic cancer sw480 were established.The Tian Ma Granules group, the blank control group, the 5-Fu group and the combined medication group were set up. The number of apoptoic cells and gene expression of wpP53 and bcl-2 were quantitatively detected and compared by the application of pathomorphology, immunohistochemistry and image analysis technique.Results:Tian Ma Granules could inhibit the growth of trans-Plantation tumor. The weight and the volume of the tumor were lower than that in the blank group(p<0.05).That in the combined medication group were the lowest. That in the 5-Fu group was lower that in the Tian Ma Granules group(p<0.05).The number of apoptotic cells in the Tian Ma Granules group were more than that in the blank group(p<0.05),but were lower than that in the 5-Fu group(p<0.05).That in the combined group were the most. On the contrary, the wpP53 and the bcl-2 count in the blank group were the most (p<0.05).That in the combined group were the lest(p<0.05).The Tian Ma Granules group s were more than that of the 5-Fu group(p<0.05).Conclusion:The Tian Ma granules could inhibit cell proliferation of human colonic cancer, and could induce the apoptosis of the human conlonic cancer.The function of inducing cell apoptosis maybe correlated with its depression of level wpP53 and bcl-2.
