Objective:Cervical cancer is one of the most common malignant diseases in the women worldwide.,according to WHO statistics,new case of cervical cancer each year,90 percent from developing countries,cervical cancer incidence and a young trend.Cervical cancer and other tumors,the incidence and development is a multi-gene,multi-step process.Research shows that the inactivation of tumor suppressor gene is an important solid tumors in the biological basis.Many scholars believe that,DNA methylation is a tumor suppressor gene inactivation of the main mechanisms.Runx3 gene is a newly discovered tumor suppressor gene,the growth of gastric cancer cells significantly inhibited.Transforming growth factorβ(TGF-β) is a kind of multi-functional activity of peptide cytokines,in regulating cell growth and differentiation,transfer,apoptosis and mesenchymal synthesis and immune suppression,and other aspects play an important role,and its main the secretion and through the next two ways role in the secretion of target cells,was considered an important growth factor in the negative regulation.Study found that RUNX3 expressiOn can lead to silence after the TGF-βsignaling disorder,caused TGF-βdeactivation signal,triggering apoptosis obstacles,leading to tumor formation. TGF-βis in high expression in a variety of malignant tumors.TGF-βthe overexpression of cervical cancer is also closely related to the degree of malignant. Currently RUNX3 and TGF-βin the coordination of cervical cancer was less reported, this study use immunohistochenistry detection of cervical cancer and precancerous lesions in RUNX3 gene,TGF-βexpression,to investigate the RUNX3 gene,TGF-βin cervical cancer,the role of development,to seek new methods of prevention and treatment targets.Materials and methods:Specimens were obtained from 45 patients with cervical carcinoma(CSCC),63 patients with cervical intraepithelial neoplasia(CIN) and 33 patients with normal cervix,which were collected from the department of pathology of The Third Affiliated Hospital of Zhengzhou University,from May 2004 to December 2006,45 patients with cervical carcinoma were all conformed by pathologically,the mean age was 36~80 years for male with age range of 58.6±18.5;The mean age was 29~72 years for CIN with age range of 41.14;and 35~55 years for normal cervix with age range of 42.01.All the surgically resected cervical carcinoma specimen were processed for routine histology and immunohistochemistry.All the patients were not treated either by chemotherapy or by radiotherapy before operation.All surgically resected specimens were fixed with 85%ethanol,paraffin embedded,and serially sectioned for histopathological diagnosis and immunohistochemical analysis. Immunohistochemistry(SP method) was applied to determine the expression of RUNX3 and TGF-βproteins.The data was analyzed by SPSS10.0 statistical software. Theχ2 test was used for the statistics,(α<0.05) was considered significant.Results:1.The expression of RUNX3 protein significant decreaded in normal cervical tissue,cervical intraepithelial neoplasia and squamous cell carcinoma of the cervis tissues was 90.91%,69.85%and 44.44%;There were significant difference between normal cervical tissue with squamous cell carcinoma of the cervis tissues,CIN and squamous cell carcinoma of the cervix tissues(P<0.05).From G1 to G2 and G3 differentiation cervical cancer,the positive immnuostaining rates for RUNX3 decreased,the positive rates were 53.85%,50.00%and 18.18%,respectively,there were no significant differences between well and moderate group(P>0.05);The RUNX3 expression in each group cells showed the positive rate had a significant different(P<0.05),relating to classification and pathology.2.Form normal cervical tissues,cervical intraepithelial neoplasia and squamous cell carcinoma of the cervix tissues the TGF-βprotein significant expression was 6.00%,41.27%and 71.11%,respectively,there were difference between normal cervical tissues with squamous cell carcinoma of the cervix tissues,CIN and squamous cell carcinoma of the cervix tissues(P<0.05);From G1 to G2 and G3 differentiation cervical cancer,the positive immnuostaining rates for TGF-βincreased, the positive rates were 61.54%,75.00%and 90.91%,respectively,there were no significant differences between well and moderate group(P>0.05);The TGF-βexpression in each group cells showed the positive rate had a significant different (P<0.05),relating to classification and pathology.3.RUNX3 protein expression in squamous cell carcinoma of the cervix in the deletion group,TGF-βpositive case was11,RUNX3 expression positive group,TGF-βpositive case was 21,there was significantly negatively relation between the RUNX3 and the TGF-β(P<0.05).Conclusion:1.The expression of RUNX3 protein significant decreaded in normal cervical tissue, cervical intracpithelial neoplasia and squamous cell carcinoma of the cervix tissues by turn.The RUNX3 protein expression deletion relate the incidence and development of squamous cell carcinoma of the cervix2.The expression of TGF-βprotein significant increased in normal cervical tissues, cervical intracpithelial neoplasia and squamous cell carcinoma of the cervix tissues by turn.The abnormal expression of TGF-βis closely related to the development process of squamous cell carcinoma of the cervix3.There was significantly negatively relation between the RUNX3 and the TGF-β, Runx3 gene ia a suppressor gene,The expression of RUNX3 protein deletion relate the incidence the CSCC,but TGF-βprotein expression increasing relate the development the CSCC.
