| Background and AimsAging is a kind of biological processes caused by a multiplicity factors interaction of genetic, internal and external environment ,it is the evolution of morphological and features accordance with the discipline occur in whole organs, tissues, cells in life cycle,and register as a series of change and disorder of systemic, progressive and decline shows caused by aging. Aging on the human body is an unfailing hot topic. Ever since a long time ago, there have been continuous explorations and researches on the causes and mechanisms of aging on the human body, but remains a mystery not yet fully solved so far. To explore the occurred mechanism of aging is an old question, but also a new field of scientific research. In recent years, with the rapid development of Chinese medicine, and the prevalent of advocate natural healthy concept ,using the Chinese medicine or pharmaceutical/food ingredients for anti-aging have good prospects.Henan is a huge geographical province, with a particular location and a varied topography, both the North and the South region plant resources rich in the North-South staggered compatible climate. Chinese date, honeysuckle and the four Huaiyao are abundant and high quality, the study of their effects is conductive to the development of the local economy.Take the whole animal's serum after a period of drug-feeding, the drug-containing serum in place of traditional Chinese medicine extracts in vitro experimental methods, called Serum pharmacology of Chinese medicine. The study has been carried out systematically, since China's State Administration of Traditional Chinese Medicine legislated the title in 1991. The advantage of serum pharmacology of Chinese medicine: reflects the compound series effect in the body; overcome the direct interference of their physical and chemical properties in the experimental study. Just like other organisms, cells have certain a life. Normal diploid fibroblasts have a limited number of the split in vitro conditions, and then they come into a irreversible standstill of proliferation, that is, aging of copying. The cell proliferation ability in vitro to reflect to the aging condition of the cells internal. Therefore Human Diploid Fibroblast Cells is recognized as a model of cell senescence study.Methods1. Extract Preparation and Detection: Add alcohol to the supernatant with fat free of hot water extractions of the Chinese date, honeysuckle, Chinese yam, chrysanthemum, and rehmanniae, and then got the water extractions resp. after the vacuum concentration. The composition analysises of aqueous extract were carried out with comparison to a specification curve. Then test the anti-free radical capacity of the aqueous extracts with the method of pyrogallol autoxidation.2. Preparation of serum with drugs: Divided the female Japanese big-ear white rabbits into two groups as following: drug groups, and control group. Drug group animals got the moderate aqueous extracts orally; meanwhile the control group animals were given sterile drinking water equivalent. Obtain the blood samples after the last dose, separation of serum, filtration after inactivated, and then reserve in -70°C refrigerator.3. Cell culture and indicators detection: Divided grops in the human embryonic lung fibroblast cell line 2BS (from the age of 20 and 40)as follows: control group, serum control group, the drug groups, cultivated four consecutive post-intervention for the latter experiment. The cells of all the groups were carried out the cell cycle and apoptosis determination; cell supernatants were carried out the antioxidant capacities.4. Statistical analysis: The numerical data obtained by experimental using the SPSS12.0 software for statistical analysis, all test data are expressed (x|-)±s. For normal data, using analysis of variance, the comparison between samples uses the q test. As for the non-normal distribution of data using rank sum test, between two groups of samples compared used Rank-based testing of M. Standard testα= 0.05.Results1. Through the Chinese date, Chinese yam, and yellow, honeysuckle, chrysanthemum medicine for five unilateral hot water extraction, respectively, by its aqueous extract, aqueous extract of the composition analysis carried out in the five water objects indicated that the total sugar content in water extract of Chinese date is the highest (43.20±0.46), total sugar content of water extract of chrysanthemum is the lowest (20.03±0.49).The aqueous extract honeysuckle has the highest total flavonoids (15.63±0.35), and the Rehmannia Flavonoids has the lowest one(1.97±0.25). Aqueous extract of the five in vitro determination of anti-free radical capacity, shows that removal ability of superoxide anion radical in chronological order is: Chinese date> Chinese yam>Dihuang> honeysuckle> chrysanthemum, also the results indicated that the inhibition rate increased gradually with the concentration increase.2. Five Chinese medicine extract can promote the proliferation of senescent cells; increase superoxide dismutase activity of SOD, reduce lipid peroxide content of MDA, to improve the metabolism of free radicals. And the extracts of Chinese date and Chinese yem have the effect of the most obvious, the one of chrysanthemum have the least.Conclusion1. The drugs contain polysaccharides, flavonoids and other bioactive ingredients, combined with previous studies show that these ingredients have the function of antioxidant and anti-free radical, a variety of substances have obvious synergy.2. The effects of anti-aging of water extract on the 2BS cells have shown that aqueous extract of each group has anti-oxidation, free radical scavenging, and enhance the body's ability to anti-free radicals, within all the groups, the aqueous extracts of Chinese date and Chinese yem are the best. Combination of the first part of the study, it suggests that aqueous extract has antioxidant capacity, and its anti-freeradical is relevant to polysaccharides and flavonoids. |
