| The objective is to investigate the influences of water extracts from Peucedanumpraeruptorm Dunn on the adriamycin pharmacokinetics in rats.In section one,water extracts from Peucedanum praeruptorm Dunn is prepared and the totalcomadin is determined. Reference to records the method of medical material extraction isadopted. The water extraction result is 16%.Adopt ultraviolet spectrophotometry to determinthe contents of total comadin in Peucedanum praeruptorm Dunn.The experimental resultindicates that the contents of total comadin is 3.04%.In section two,the HPLC method is used to determin adriamycin plasma concentrations.Pre-disposal method is established of blood plasm sample. The extraction method was chosento pretreat plasma samples before HPLC analysis. The analytical column was DiamonsilODS(4.6mm×200mm,5μm). A mobile phase of 0.01 moL·L-1 ammonium dihydrogenphosphate and methyl cyanide (77:23,Ph4) was run at a rate of 1.0 ml/min and the sample sizeis 20μL,column temperature at room temperature. The calibration curve was linear within therange of 0.0156~10.0μg·mL-1 for adriamycin and the detection limit was 3.1ng (S/N≥10),both of the intra-day and inter-day precision were less than 10.0%.The stability and recoveryrate consistent with analysis requirement.In section three,water extracts from Peucedanum praeruptorm Dunn impact on adriamycinpharmacokinetics in rat is studied. The rats were divided into A1,A2,A3,B1,B2,B3,C1,C2,C3,D groups randomly,four rats in each group. The high performance liquidchromatography(HPLC)method measuring adriamycine concentration in Plasma was used tomeasure and comparethe pharmacokinetic parameters among the groups.The phannaeokineticsparameters were calculatedas followed:①Compare group A with the group that injectadriamycin singlely,water extracts from Peucedanum praeruptorm Dunn make the AUC ofadriamycin accrete notablely,and velocity of adriamycin eliminat from central compartment toperimeter compartment step down.,also,the rate of elimination constant decrease and theother parameters is similar to adriamycin.②Parameters of the other groups is similar toadriamycin,there is no statistical significance.In section four,water extracts from Peucedanum praeruptorm Dunn impact on adriamycindisposition in rat is studied.Use the same HPLC condition,the calibration curve was linearwithin the range of 0.1~20μg·mL-1 for adriamycin.The stability and recovery rate consistentwith analysis requirement,both of the intra-day and inter-day precision were less than 10.0%.In the experiment,the 32 rats were divided intoA,B groups randomly,16 rats in each group.The high performance liquid chromatography(HPLC)method was used to measure adriamycine concentration in Plasma,the result show that when water extracts from Peucedanumpraeruptorm Dunn and adriamycin used together,adriamycine concentration in Plasma raises,especially the heart,liver and kidney. |
