Shugan Yiyang Capsule On The Expression Of Nitric Oxide Synthase Isoforms Arterial Erectile Dysfunction In Rats

Objective:Components of Shuganyiyang capsule have good effects on invigorating the circulation of blood and Qi, nourishing kidney, and improving erectile dysfunction. Shuganyiyang capsule also shows good clinical effect on curing ED. The objective of the article is to reveal the mechanism of how Shuganyiyang capsule cures ED by changing NOS subtype molecular expression of cavernous body of penis.Methods:1. Choose 3-month-old adult male SD rats (weight250-300g).2. After raising the rats for 3 days, based on literature, use internal iliac artery ligation method to create model rats with vascular erectile dysfunction. Then, observe for 2 weeks and choose the rats having no complications as models.3. Divide experimental rats into 4 groups,10 for each group:SGYY low-dosage group, SGYY high-dosage group, Sildenafil group, model group. Meanwhile, keep 10 normal rats as control group. Feed SGYY low-dosage group with, SGYY high-dosage group with 1g/kg/d, Silenafil group with 0.5mg/kg/d, model group and control group with distilled water for 30 days.4.1 week after finishing dosing, conduct ICP test and compare erectile pressure of cavernous body of penis between each group.5. Nitrogen freeze the tissue fluid of cavernous body of penis and conduct PCR and western blot to test the activity of nNOS, eNOS, iNOS.Conclusion:PCR showed Shuganyiyang capsule could enhance the expression of NOS subtype molecular mRNA, and enhance the expression of iNOS, eNOS, nNOS mRNA in cavernous body. Western Blot proved Shuganyiyang capsule could significantly enhance nNOS, iNOS, eNOS protein expression in cavernous body. High-dosage showed higher effectiveness than low-dosage. In summary, this article proved the effectiveness of Shuganyiyang capsule on curing vascular ED. Meanwhile, revealed the mechanism of how Shuganyiyang capsule cures vascular ED by testing mRNA and protein expression.
