Objective:To study the expression of the matrix metalloproteinases-9 (MMP-9) and the role in the formation of aortic aneurysm.To assess the traditional Chinese medicine Si Miao Yong An soup treat the aortic aneurysm by control the expression of MMP-9.Materials and methods:36 Wistar rats (18male and 18females), 24 of which are used to set up abdominal aortic aneurysm elastase perfusion models. According to randomized controlled searching form, the rats are divided into 3 groups (12 rat in each group, 6 males and 6 females): experiment (Si Miao Yong an soup) group, normal control group, model control group.Elastinase was perfused into the experiment group and model control group abdominal aortae to construct AAA animal models. Experiment (Si miao Yong an soup) group are administered 3ml Si miao Yong an soup decoction, model control group are infused respectively the same dose of saline. 3 weeks after the perfusion, the expansion rate of arterial and to observe changes in arterial wall histology is calculated. The expression of the MMP-9 in experiment groups and control groups are detected by immunohistochemistry and in situ hybridization .Results:1. 3 weeks after the perfusion, the expansion rate of arteris is 54.9﹪±2.9﹪in experiment (Si miao Yong an soup ) group,107.8﹪±6.1﹪in the model of control group,respectively (P<0.05). Arteries do not expand in normal control group.2. The results of immunohistochemistry showed that 3 weeks after the perfusion, the expression of the MMP-9 in experiment (Si miao Yong an soup) group is weak positive. The expression of the MMP-9 in the model of control group is strongly positive. The differences among groups were statistically significant ,(P<0.05). 3. The results of in situ hybridization found the expression of the MMP-9 mRNA cells in experiment (Si miao Yong an soup) group is weak positive, The expression of the MMP-9 mRNA in the model of control group is strongly positive, MMP-9 mRNA expression is negative in the normal control group. The differences among groups were statistically significant ,(P<0.05).Conclusion:Excessive expression of MMP-9 was a principal element in the development and progression of AAA. MMP-9 expression can be reduced by the traditional Chinese medicine Si miao Yong an soup, which can inhibiting the formation of abdominal aortic aneurysm in rat.
