| or the purpose of systematic quality evaluation of Semen Armeniacae Amarum, Semen Persicae and their prescriptions, the following studies were carried out.1.A reversed-phase high performance liquid chromatography (RP-IIPLC) method was developed for the assay of aniygdalin in both Semen Armeniacae Amarum and Semen Persicae, which is an active and slightly toxic constituent. The quantitative analysis was performed on a Hypersil ODS column with a mobile solvent of methanol and water (1 :4,V/V), with method showing good recoveries of 96.1% and 98.7% respectively, and the linear range being O.1005挆1.OO5mgImL (r0.9999). There was a significant difference in amygdalin content between the crude drugs and the processed ones, which means that the quality of drugs were closely related with the processing method. Moreover, two TLC systems were used in analysis of the drugs with which clearer chromatograms with better separation and sensitivity were obtained than with method described in the Chinese Pharmacopoeia. Furthermore, a systematic quality evaluation of the two crude drugs from the different-3-markets was shown by a radar diagram based on the following five parameters: amygdalin contents, the amounts of lipid, ethanol-soluble extract, total ash and acid-insoluble ash.2.The amygdalin contents in the 7 Semen Armeniacae Amarum or Semen Persicae梒ontaining Traditional Chinese Prescriptions and 5 relevantcommercial prescriptions were determined by using HPLC, in whichtrecoveries were 96.&-l00.6% and the linear range was0.1 104æ¢.84OmgImL (r =0.9998) by using metbanol-acetonitrile-waterglacial acetic acid (1:1: 16:0.02,VIV) as the mobile phase. Because of a good accuracy, adaptability and a simple process, it will be expected to be a standardized method for the quantitative analysis of aniygdalincontaining prescriptions.3.This investigation on the amygdalin availability in Traditional Chinese Prescriptions suggested that it is also closely related with the processed properties of the material crude drugs, with the highest availability resulted from the boiled- parched ones. |
