Objective: To probe into the action mechanism of Shugan Ruanjiangtang(SGRJT) on immune hepatic fibrosis in this experiment effect of SGRJT on immune hepatic fibrosis rats were observed from histomorphology , immunohistochemistry and molecular biology aspects .Methods: Fibrosis were induced by the pig's serum , SGRJT was used as therapeutic group and colchicine as control. Hyaluronic acid(HA), Laminin(LN), type III procollagen(PCIII) of serum were measured by means of Radio-immunity. Deposition of type III collagen , Transforming growth factor 1 (TGF 1), Demin(Dm) were studied in liver sections by immunohistochemical technique using special antibody. The hepatic contents of type III mRNA were determined by in situ hybridization technique.Results: Serum levels of HA, LN, PCIII in treatment group were less than those in control group (p<0. 05). SGRJT markedly decrease the deposition of type HI collagen , TGF1, Dm in liver as well as the expression of type III collagen mRNA .Coclusion: SGRJT has an antifibrotic action and could be significant and applicable for liver fibrosis. The mechanism may be related to inhibition of hepatic stellate cell (HSC) activation^ expressions of TGF i and deposition of collagen. |
