To explore the relations of PBR and liver injure in schistosomiasis and to illuminate the mechanism of huangqi protecting liver and against liver fibrosis. Method1. We choose fifty-eight mice randomly from seventy-six BALB/C male mice, which weight from sixteen kilograms to eighteen kilograms. Each mouse is artificially infected with 25 Japanese schistosome in its abdomen.2. We mark the eighteen uninfected healthy mice as group A and feed them on normal feedstuff. Six mice are anatomized respectively in the eighth, twelve and twentieth week and their livers are natural contrast.3. We divide the infected mice into three groups, namely infected group B, huangqi treated group C and 0.9%Nacl contrast group D. Thirty mice in group B are fed under the same condition and ten of them are anatomized respectively in the eighth, twelfth and twentieth week, their livers and will be used later. Sixteen mice in group C are fed under the same condition and each is injected with 0.2 ML huangqi injection into its abdomen everyday since the eighth week. Eight mice are anatomized respectively in the twelfth and twentieth week; their livers will be used later. Twelve mice in group D are fed under the same condition and each is injected with 0.2 ML 0.9%Nacl into its abdomen everyday since the eighth week. Six mice are anatomized respectively in the twelfth and twentieth week; their livers will be used later.4. We fix part of livers of the four groups with 10 percent neutral formaldehyde and chop them in 5 UM continuous slices after embedment with olefin. Then we observe these slices from the angle of histopathology. Part of them are place in the -70 癈 refrigerator and they are used to determine the quantity of PBR-mRNA through PCR(FQ-PCR).Results1. Histopathology observationIn the eighth week, egg granuloma pathological changes in the infected mice's livers come into being, fibrous connective tissue hyperplasia in C, inflammation reaction is strong. Part of granuloma centre begin to die and form acidophilic abscess; In the twelfth week, fiber hyperplasia is easily seen in portal area and around egg granuloma ,many chronic egg granuloma focus have formed fibrosis; In the twentieth week, a mass of fiber scar focus could be seen in portal area. Hepatic sinusoid is oppressed. But the tissue modality and structure of the mice's livers in the natural contrast are normal.2. PBRIn the eighth week, the PBR of infected mice's livers clearly increases comparing with that of the normal ones( P < 0.05 ). In the twelfth and twentieth week, the PBR of infected mice's livers also clearly increases comparing with that of the normal ones (P< 0.01 ) . The PBR of the infected livers in different term increases gradually (P < 0.01 ), but that of the uninfected livers in different term almost remains the same. (P > 0.05 ) .The liver's PBR of the huangqi treated group is less than that of the mice infected at the same term( P < 0.01 ). The livers' PBR of the 0.9%Nacl contrast group is almost the same with that of the mice infected at the same term( P > 0.01). The liver's PBR of the huangqi treated group is less than that of the physiology brine contrast group at the same term ( P < 0.01 ) . The livers' PBR of the huangqi treated group is almost the same with that of the natural contrast group at the same term ( P > 0.05 ) . Conclusion1. PBR of the livers increases distinctly in the course of liver injure of the mice infected with schistosomiasis. And PBR expression goes with the degree of liver injure in schistosomiasis.2.
