Objectives: To study the effect of the guaianolide compound, Thapsigargin (TG), on the intracellular calcium in MCF-7 cells.Methods: Concentrations of intracellular calcium in MCF-7 cells were measured by calcium sensitive fluorescence indicator Fura-2/AM techniqe in vitro with different does of TG.Results: The concentration of intracellular Calcium in MCF-7 cells was increased obviously with the adding up of TG. We found it was 242.48+10.116nmol/L in control cells. When the concentration of TG was 50nM,100nM and 250nM respectively,the intracellular calcium was 590.12+9.75 nM, 692.85+13.26 nM and 803.50+6.68 nM accordingly. Values of P in Newman- keuls were all less than 0.01.Conclusions: guaianolide compound, TG, may raise the concentration of intracellular calcium in MCF-7 cells definitely.
