| The uterine leiomyoma is one of the most common benign tumor in women, occurring in 20% to 25% of reproductive period over 30 years older, and is the main cause of hysterectomies. The real patho-genesis is unknown, but that the development and growth of the uter-ine myoma have relationship with stimulation of the steroid hormones secreted by the ovaries may be decided. The recent evidence has dem-onstrated that the production of uterine myoma do not come from one of kinds of causes, whereas many detail points may be promote leiomyo-ma cell growth. The polypeptide hormones, insulin - like growth fac-tor - I and II (IGF - I , IGF - II ) have possible roles in the growth of uterine leiomyoma. These growth factors may function as autocrine and paracrine mediators of sex steroid - induced cell growth and differ-entiation by altering the onco gene and the tumor suppressor gene. Be-cause the insulin - like growth factors are potential mitogens, and there are increased numbers of binding sites for insulin - like growth factor in leiomyoma cell.To investigate the correlation between IGF - I , IGF - II and the development and growth leiomyoma, as well as the correlation between expressionof IGF - I , IGF - II and menstrual calender, and provide experimental data to the growth machinism and treatment of leiomyo-ma. we observed the effect of IGF - I , IGF ?II on human uterine leiomyoma.Materials and MethodsMaterials. Tissue samples of normal myometrium and uterine leio-myoma were obtained at hysterectomy from thirty reproductive women who had not received exogenous hormonal or gonadotropin releasing hormone agonist therapy in three weeks before their surgery . Sixteen of the patients were in the proliferative phase, ten were in the secretory phase, one were in menopausal phase, and three were in other states, as confirmed by menstrual calender and histologic datings. Tissue specimens were examined by gynecol apathologist. We colleged the samples of uterine leiomyoma nucleus and their own normal myometri-um that were over 2 centimeter from their wrapping membrane of leio-myoma. All specimens colleged after operation were placed in Eppen-dorf tube, stored in refrigerator at -70? C and waited to be used.Reagents. Trizol and reverse transcription kit were purchased from TaKaRa Inc. The primers of insulin - like growth factor - I , insulin - like growth factor - H and β - actin mRNA were synthesized by Beijing AoKe biological Inc.RT - PCR and DNA ladder reagents were proviced by the con-genital malformation lab of the second clinical college of China Medi-cal University.Methods.
