| Purpose:This experiment rests on the Traditional Chinese Medicine's principle of treatment according to the syndrome differetiation, the concept of wholism and finding its root when treating a disease, by testing the serum sexual hormone's levels of chloasma patients before and after the treatment as well as the control group of normal healthy women, meanwhile, detecting the influence of ZengXiaoHaiTangMei to the activity of tyrosinase in vitro using mushroom-tyrosinase dopa-rate oxidation process, the purpose is to expound ZengXiaoHaiTangMei's mechanism of prevention and treatment to chloasma,and to explore the etiology and pathogenesis of chloasma. Method: 60 chloasma patients are divided into chloasma-treated group and positive control group randomly, and each group 30 cases. Then choosing 30 other healthy women as healty control group, compared with chloasma treated group in age, P>0. 05. Give the chloasma treated group ZengXiaoHaiTangMei capsules, 0. 72g, 3 times/daily, orally. Give the positive control group BaiXiaoDan pill ,8g,3 times/daily. Both of the two groups take medicine one month being a therapeutic course. Then evaluate the therapeutic effects after two continous therapeutic courses. The serum sexual hormone (E2, P,T) levels are measured both in the patient group before and after treatment and in the healthy group testing time same as patient group , the method, of which radioimmuuoassay method. At the same time, the influence of the compound ZengXiaoHaiTangMei and each of its composition to the activity of tyrosinase are test in vitro. All the data use the statistical package for the Social Science, and make analysis of variance and t-test.Result: Compared with the healthy control group, the levels of estrogen (E2) and progesterone (P) of the choasma treated group are much higher(P<0. 05) , while its level of androgen(T) is much lower (P<0. 05). After treatment, the level of estrogen (E2) and progesterone (P) of the chloasma treated group declined obviously(P<0.05),while level of androgen (T) raised markedly(P<0. 05). However the positive control group had no obvious change compared with pre-treatment (P>0. 05). The alchoholic extract of ZengXiaoHaiTangMei makes the activity of tyrosinase declined(P<0.01), while aqueous extract makes it increased(P<0.01).Conclusion: These results Suggest that the occurrence of chloasma in female is related to endocrine-metabolic disorder. Because theestrogen secretion results in metabolic bloc of melanin. The latter leads to large amount of deposit of melauoeyte on epidermal cells. The reason why ZengXiaoHaiTangMei can treat dhloasma effectively has experimental basis. It has the functions of regulating endocrine , balancing hormones level and inhibiting the activity of tyrosinase. |
