Purpose: In order to observe the effects of Yi-Qi-Huo-Xue-Jie-Du -Fang to proliferation and activity of enzyme of lung cancer cell and explain the mechanism of its resisting-cancer function, we established the mouse model of the Lewis lung cancer and cultured the human pneumoadenocarcinoma cell A549 in vitro. The result can provide reliable evidence to the application of Yi-Qi-Huo-Xue -Jie-Du-Fang in clinic on treating the lung cancer. method: The experiment included the vivo part and the vitro part: In vivo part, we chose the cell strain of Lewis lung cancer mouse, inoculated the cell strain subcutaneously. After building the tumor animal model successfully, we divided the animal into three groups: Saline control group; cisplat in chemiotherapy group; Yi-Qi-Huo-Xue -Jie-Du-Fang treating group. Using the method of light microscope (LM) and electron microscope (EM) to observe the morphology. In vitro part, we cultured the cell strain of the human pneumoadenocarcinoma cell A549, MTT method was used to assay the proliferation of human pneumoadenocarcinoma cell A549caused by serum contained Chinese herbs. The enzyme cytochemistry was used to detect the activity of G-6-Pase and Mg2+-ATPase of the lung cancer cell.result: In vivo part: among 40 Lewis lung cancer mice in experimental group, the episode mice of cisplatin chemiotherapy group is 14. Yi-Qi-Huo-Xue-Jie-Du-Fang treating group has 16 episode mice, while the control group has 20 samples. To be compared with the control group, the experimental group has following characteristics: the tumor ball and the cell volum are smaller, the hetero-nature is not evident, and the pathological karyokinesis and interstitial blood vessel are fewer. It has more severe necrosis and infiltration of lymphocyte. Through the observation of EM, we found the Chinese herbs can activate the proliferation of lymphocyte. There are many lymphocytes between the tumor cells and the lymphocytes invade the tumor cells to destroy the tumor cells 2. In vitro part: (1) the serum contained Chinese herbs has evident inhibitory function to the human pneumoadenocarcinoma cell A549 after 24h culture. The inhibiting rate is 47. 83%. It has more obvious inhibitory action after 48h culture. The inhibitory rate is 61. 29%. The inhibitory effects have time dependence. We compared the different effects of the' human pneumoadenocarcinoma cell A549 proliferation in different culture time. The inhibitory action of 3d-2t-2h group is the best. The inhibitory rate is 66.41%. (2) Enzyme cytochemistry method showedthat the response granules of G-6-Pase and Mg2+-ATPase in the human pneumoadenocarcinoma cell A.549 become smaller after the treatment of serum contained Chinese herbs, the amount of the granules became fewer, the density was lower, some were negative. The activity of the enzyme decreased obviously.Conclusion: 1. the has the function of inhibiting the proliferation of cancer cells and activating the proliferation of lymphocytes of Lewis lung cancer mouse. 2. The serum contained Yi-Qi-Huo-Xue -Jie-Du-Fang has the function of inhibiting the proliferation of the human pneumoadenocarcinoma cell A549. 3. Using the serum contained Chinese herbs 3d-2t-2h, we think it is the reliable serum pharmacology method to research the action of Yi-Qi-Huo-Xue -Jie-Du-Fang to the human pneumoadenocarcinoma cell A549. The inhibitory function of serum contained Yi-Qi-Huo-Xue-Jie-Du-Fang to the lung cancer reacts through multipath and multiple factor. Its important mechanism of resisting cancer is inducing the activating the proliferation of lymphocytes ; inhibiting the activity of the telomerase and G-6-Pase and Mg2+-ATPase;...
