| Viral myocarditis (VMC) is a well-known disease associated with characteristic lesions, such as necrosis of myofibers, inflammatory cell infiltration and fibosis. Coxsackie virus B group can easily attack myofibers. It can not only make inflammation and necrosis of myofibers. but also induce different rate and serious of disease with different virus strains. Because maximal inflammation develops after a significant decrease in virus titer, it is believed that both immunological mechanisms and the direct cytolytic effects of viruses play an important role in acute viral myocarditis. Incidence of VMC has gradually risen since 70's. But so far, there is neither effective treatment to it, nor radical cure drugs. Traditional Chinese Medicine has an evident advantage of preventing and treating VMC, so it is an urgent task to develop a good medicine in Chinese herbs. Sheng Mai Bao Yuan juice(SMBY) is an prescription of Li GAO. It has an extra effect on palpitation, chest distress, dispnea, heart function and ECG. So an experimental study on the inhibitory activity of SMBY against coxsackie virus B group was carried out in mice, so that il can give experimental basis of treatment of viral myocarditis and dilated cardiomyopathy.Methods: In the experiment, all the experimental animals were randomly divided into six groups, namely ,the normal control group, the pathologic control group, the Ribavirin group, the high dose SMBY group, the middle dose group, and the low dose group. Every animal was induced by means of injecting coxsackie Bjm virus (0.lml/per mouse)into abdominal cavity except the normal control group.SMBY groups fed with 20g/kg,10g/kg and 5g/kg per day respectively. In each group, mice were randomly killed from the 5th day to the 9th day .Our purposes is to research the effect of SMBY on the death rate, the pathological changes of myocardium and the expression levels of TNF-α mRNA and 1FN-Y mRNA in mice infected by coxsackie B3m Virus weight. TNF-α mRNA and IFN- # mRNA were tested by the. way of reversetranscription polymerase chain reaction(RT-PCR).Results: Symptoms, such as Pilo-erection, lassitude, loss of body weight and loose stool, occurred on 3rd day. The death of mice occurred on 4th day, reached climax from 7th day to 9th day. Compared with the pathologic control group, symptoms were relieved and the rate of deathwas significantly decreased in the high dose SMBY group. NO pathologic phenomena occurred in the normal control group. The pathological findings of myocardium of the mice in the high dose SMBY group decreased apparently compared with those of other groups. The expression of TNF- α mRNA in VMC mice increased markedly and was higher than that in the high dose SMBY group. But the level of 1FN- γ mRNA expressed in VMC mice was lower than that in the high dose SMBY group.Conclusions: Results showed that the SMBY can decrease the death rate, relieve the pathological findings, promote inducing IFN-γ and inhibit TNF-α .SMBY had therapeutic effect on VMC.At the end, combining the theories of TCM with modern researches, the thesis sets forth the foundation of SMBY's constructing formulate and using medicine.
