| ObjectiveTo explore the etiological factor and effect of replenishing kidney-essence, remove phlegm and promoting blood circulation therapy on treating Alzheimer's disease(AD) in theory. And to observe the effects of "WEN-DAN improvement decoction"(WDID) on learning memory and cholinergic system for analyzing the mechanism of action of WDID for AD. Methods15-month Wistar rats were randomly divided into normal group, blank group, AD model group, piracetam treated group, high-dose and low-dose of WDID group, then induced by injection of basal nucleus of Meynert (BNM)with ibotenic acid(IBO) to make AD model. After treating for four weeks, Morris water maze was used to observe the ability of space learning memory and radioimmunoassay was used to assay the activity of acetylcholine(Ach),and immunohistochemistry method was used to detect the content of the induction nitricoxide synthase (iNOS ) and the nerve growth factor (NGF) in brain. Results1 In the behavior experiment, not all the ability of learning gain and the competence of information reserve ,the significant differences were observed between the model, piracetam treated and normal group (p<0.01); also between the high-,low-treated group of WDID and piracetam treated(p<0.01).2 The activity of Ach was lower in AD model rats than that of normal rats obviously (p<0.01). The remarkably improved rates of the three medicine groups(p<0.01).the high-,low-treated group of WDID were 1.6 and 1.9 times than the model group respectively. There were significant differences between the high-,low-treated group of WDID and the piracetam treated group (P<0.05-0.01) .And the activity of Ach in the high-treated group of WDID was near to the level of normal group(p<0.01).3 The marks of the iNOS concerned on AD mode of was higher than the other groups . The remarkably decreased rates of the three medicine groups(p<0.01). there were not significant differences between the low-treated group of WDID and piracetam treated group (P > 0.05). the high-treated group of WDID was superior to the low-treated group of WDID and piracetam treated group(p<0.01).4 To the high-,low-treated group of WDID group and the piracetam treatedgroup, the decrease of NGF in model group had highly remarkable differences(P0.01).NGF content of the high-,low-treated group of WDID were6.5 and 4.5 times than that of the model group respectively, while the high -treated group of WDID group rised highly significantly (P0.01).And it did arrive the level of normal group(P<0.01).ConclusionKidney-essence weak,phlegm and blood stasis is the basic etiological factor for AD,and replenishing kidney-essence, removing phlegm and promoting blood circulation therapy is operative on treating AD. WDID possesses a certain improvement of the Ach and NGF content, decreased the iNOS content for AD model rats, consequently increase the learning memory level .Obviously, the WDID groups were superior to the piracetam group. And the high-treated group of WDID was more significant. |
