objective: to induce EAE model in un-sensitive Wistar rats with different dose Mycobacterium bovis bacillus Calmette-Guerin(M.bovis BCG), and to observe the kintic change of MCP-lmRNA expression, as well as to study the immunosuppressive effects of triptolide (TRD, a purified component from a traditional Chinese herb, Triptery gium wilfordii Hook F) co-treatment with dexamethasone (DXM).Methods: first, Wistar rats are divided randomly into EAE1, EAE2, EAE3,EAE4 for animal model; then, the normal control group, the another control group, EAE group for the kintic change of MCP-lmRNA expression; last, GPSCH, CFA and PT-immunized Wistar rats were divided into six groups for the immunosuppressive effects of the medicine. The brains and spinal cords were removed for HE staining and in situ hybridizations(ISH) staining, results: (1) EAE3 and EAE4 is higher in the incidence (p<0.05) ,weight change, clinical score of EAE, the expression of MCP-lmRNA. (2)In the kintic changes of MCP-lmRNA, the expression of MCP-lmRNA are correlation with clinical signs of disease.(3)the TRD3,the DXM,the TRD co-treatment with DXM group are lower in incidence of EAE , the development of clinical score, the expression of MCP-1 mRNA. ConcIusion:(1)The degree of M.bovis BCG can influence incidence of disease.(2)MCP-l appear the key function during pathology process of EAE.(3)TRD and DXM all have treat effect.(4)TRD cotreatment with DXM can decrease the dose of medicine,but rise same treatment effect.
