Objective: To study the changes of IgE, IL-4 and IFN-y in serum and pulmonary tissue homogenate of asthmatic guinea pigs , search for the relationship between IFN- /IL-4 and IgE and the effects of Scoparone on them. Methods: To divide animals into three groups: control, asthma and scoparone treatment groups. Choose the model guinea pigs of asthma sensitized with OA, observe the changes of IgE, IL-4 and IFN-y in serum and pulmonary tissue homogenate of asthmatic guinea pigs by means of chemolumi nescence ,radio immunoassay , enzyme linked immunoabsordent assay (ELISA). search for the relationship between IFN- Y /IL-4 and IgE and its effect on pathogenesis of asthma , and the effects of Scoparone on them. Results:IgE and IL-4 in serum and pulmonary tissue homogenate of asthmatic guinea pigs obviously increase (P<0.05), IFN-y obviously decrease (P<0. 05). Scoparone can obviously decrease the content of IgE and IL-4, increase the content of IFN-y (P<0. 05)in serum and pulmonary tissue homogenate of asthmatic guinea pigs. According to the analysis of relationship :There were a significant correlation between IL-4, IFN-y, IFN-y/IL-4 and IgE including a negative correlation between IFN- y /IL-4 and IgE (in serum P<0. 05; in lung tissue P<0. 01). The result indicates that the disorder of IFN-y/IL-4 and increasing level of IgE may play an important role inthe pathogenesis of asthma. Conclusions: Scoparone may modulate the balance between IL-4 and INF-γ to inhibit the abnormal synthesis of IgE in asthmatic guinea pigs.
