Objective:To study the effect of "shuanggu Yitong" acupuncture therapy on the bone mass density (BMD), serum immunoglobulin (Ig) and the expression level of receptor activator of NF-kB ligand (RANKL) mRNA in bone tissue of the experimental rats with postmenopausal osteoporosis (PMO). To supply experimental evidence approving acupuncture can prevent and treat osteoporosis. Methods:Choosing female SD rats aged 6-10 months then resecting double ovaries and feeding them for 90 days in order to make the model of experimental osteoporosis. Then, dividing them into 5 groups such as control group, regular acupoints applying group, Shuanggu group, Yitong group, Shuanggu Yitong group, with 12 rats each group. There wasn't any therapy in control group; We applied Pishu(BL 20), Weishu(BL 21), Shenshu(BL 23), Qihaishu(BL 24) for the regular acupoints applying group; Guanyuan(RN 4), Zusanli(ST 36) for the Shuanggu group; and Shenshu(BL 23), Geshu(BL 17), Dazhu(BL 11) for the Yitong group; Guanyuan(RN 4), Zusanli(ST 36), Shenshu(BL 23), Geshu(BL 17), Dazhu(BL ll)for the Shuanggu Yitong group. Giving electroacupuncture therapy once a day, 15 minutes each time, holding the needles for 10 minutes. Then these rats rested for 1 day after consecutive therapy for 6 days. The therapy lasted for 8 weeks. Measure: (1) Using dual-energy X-ray absorptiometry to measure the rats' bone mass density (BMD) of shinbone. (2) Applying array protein system(APS) to examine serum Ig. (3) Using RT-PCR to detect the expression level of receptor activator of NF-kB ligand (RANKL) mRNA in bone tissue. Results:(1) The indexes of BMD measurement indicate: Compared with control group, in the other four groups, BMD all increased (P<0.05), BMD increased significantly in Shuanggu Yitong group (P<0.01); Compared with regular acupoits applying group, Shuanggu group, Yitong group, BMD increased in Shuanggu Yitong group (P<0.05); There are no statistic significance among regular acupoits applying group, Shuanggu group, Yitong group (P>0.05). Conclusion: Acupuncture can increase BMD of osteoporosis model rats without ovaries and "Shuanggu Yitong" acupuncture therapy superior to other regular acupoints applying acupuncture therapy.(2) The indexes of immunoglobulin (Ig) measurement indicate: serum IgA is not different significantly among 5 groups (P>0.05). Compared with control group, in the other four groups, serum IgG and IgM all increased (P<0.05), the indexes increased significantly in Shuanggu Yitong group (P<0.01); Compared with regular acupoits applying group, Shuanggu group, Yitong group, serum IgG and IgM allincreased in Shuanggu Yitong group (P<0.05); There are no statistic significance among regular acupoits applying group, Shuanggu group, Yitong group (P>0.05). Conclusion: Acupuncture can increase serum IgG and IgM of osteoporosis model rats without ovaries and "Shuanggu Yitong" acupuncture therapy superior to other regular acupoints applying acupuncture therapy.(3) RANKLmRNA' expression level measurement indicates: Compared with control group, the expression level of RANKLmRNA in all acupuncture therapy groups increased (P<0.05); The index didn't differ significantly among regular acupoints applying group, Shuanggu group, Yitong group, Shuanggu Yitong group (P>0.05). Conclusion: Acupuncture can improve the expression level of RANKLmRNA in bone tissue of osteoporosis model rats without ovaries. Final conclusion:"shuanggu Yitong" acupuncture therapy can increase BMD, serum Ig , the expression level of RANKLmRNA in bone tissue of osteoporosis model rats without ovaries, and "Shuanggu Yitong" acupuncture therapy is superior to other regular acupoints applying acupuncture therapy increasing BMD and serum Ig.
