| ObjectivesAn experimental observation was made on the basis of the therapeutic effect of Gutong capsule on rabbit models of knee osteoarthritis made by Hulth's technique. To observe the pathologic form of articular cartilage and content of SOD, HA,NO in the serum , study the effect of Gutong capsule in the treatment of osteoarthritis and histo-pathological basic of the clinical treatment. Methods1.Grouping32 rabbits were randomly divided into 4 groups: the normal group, the model group,the Gutong capsule group(the test group)and the Kanggu zengsheng capsule group (the control group), 8 for each group. The normal group does not make any handling,the model group, the test group and the control group were duplicated by Hulth's technique.2. Treatment methodThe animals in the test group were fed with Gutong capsule 0. 28g/kg ever day, and Kanggu zengsheng capsule in the control group 0. 245g/kg ever day, curing for 8 weeks continuously.3. Observation to the pathologic form of articular cartilageAfter curing for 8 weeks, all the animals were sacrificed respectively and specimens were procured from the medial condyle of femur and the medial plateau of tibia for light microscopic and transmission electron microscopic study.4. Measurement to the content of SOD, HA, NOTake the venous blood 2 ml, and centrifugation, then measure to the content of SOD, HA, NO according to operational method. Results1. The right knee joint X-ray, the normal group:the medial and lateral interspace of knee joint were homogeneity, the adumbration of the medial condyle of femur and the medial plateau of tibia were clear;the model group: the medial interspace of knee joint was narrow obviously; the control group and test group:compared with the control group, the adumbration of the medial condyle of femur and the medial plateau of tibia were neat.2.By the electron microscope, light microscope and eyes, the articular cartilage of the normal group was not broken, the articular cartilage of the model group was broken seriously,the remodeled articular cartilage were broken seriously, surface layer cartilage contain much crack, peeling off formation defection and damage area, the cartilage cell arranges the mess, four layer constructions distinguish not easily, the tide line is not complete much. The rough endopasmic reticulum degranulate, the number of the mitochondria reduced, necrosis cell broken bits therefore,cell nucleus shape anomaly, the chromatin concentrated, spread in the nucleus. and the articular cartilage of the test group's was broken less seriously than the one of model group. articular cartilage surface contain shallow cracks, the cartilage cell arranges tidy, report the pillar form, the tide mark line is complete, therefore more not mature cartilage cell,the rough endoplasmic reticulum expansion pond, the golgi complex extended, therefore the cartilage cell secretes the gum original, and appear the more egg with many sugar grain,the stability that maintain the cartilage cell the membrane, promote the cartilage cell propagate to combine to urge to have a great achievement can of the cell machine increases to living, hinting the cartilage cell resumer the normal metabolism the level. ConclusionThe experiment result showed that Gutong capsule has the clear and definite treatment effect to the articular cartilage of the osteoarthritis model domestic rabbit, backing to change to prevent the function of thearticular cartilage. Promoting the cartilage cell propagates to combine the cell machine that urge it has a great achievement and can increase living, improving the cartilaginous nourishment in joint circumstance, recovering the cartilaginous and normal physics and chemistry in joint environment to the best, the cartilage cell resumed the normal metabolism level. and relieving or slowing the cartilage degeneration.It's difficult to repair the apparently degenerative articular cartilage,but can prevent the damage from getting worse. |
