OBJECTIVE 1 Take measurements of the nucleus diameter of microemulsions containing Andrographolide to define that the diameter is less than 100nm. 2 Take measurements of the concentration of microemulsions containing Andrographolide to define how many Andrographolide are there in the microemulsions 3 Observe and study the stability of microemulsions containing Andrographolide 4 Compare the bioavailability between microemulsions containing Andrographolide and Andrographolide pil METHODS 1 The measurements of the nucleus diameter of microemulsions containing Andrographolide Stain the microemulsions containing Andrographolide with 2% Phosphotungstic acid then observe the length of the nucleus diameter directly through the TEM. 2 The measurements of the concentration of microemulsions containing Andrographolide Take the measurements of the concentration of microemulsions containing Andrographolide through HPLC 3 The research on the stability of microemulsions containing Andrographolide Take 3 batches of microemulsions containing Andrographolide. Based on the criterion of the quantities of Andrographolide and OD, observe the changes of the quantities of Andrographolide and OD in the normal temperature,4 ℃ ,-10℃,40℃,60℃,80℃ and the changes before and after centrifugalism at the rate of 3000r/min,4000r/min and 5000r/min. 4 Comparison of bioavailability between microemulsions containing Andrographolide and Andrographolide pill Microemulsions containing Andrographolide and Andrographolide pill were given orally to rabbit respectively (8mg of Andrographolide/kg of body weight). The plasma concentration of Andrographolide was estimated by HPLC after oral administration. Then make the chart of time- concentration relationship, and calculate the parameters of bioavailability. RESULTS 1 The spreading shape of microemulsions containing Andrographolide is round. The nucleus scattered well-distributed. The diameters of the nucleus range from 30nm to 100nm. 2 The quantity of microemulsions containing Andrographolide is 8mg/ml. 3 The stability of microemulsions containing Andrographolide is fine from -10℃ to 60℃.There are no obvious changes in the OD before and after centrifugalism at the rate of 3000r/min,4000r/min and 5000r/min. 4 All Andrographolide plasma levels after oral administration of microemulsions are obviously higher than those after intake of pills, the parameters of bioavailability being: Cmax(ug/ml) 0.480977 and 0.322338, Tmax(h) 0.25 and 0.5 respectively. It is indicated that the dissolution and absorption velocities of active component of Andrographolide in the microemulsions are obviously predominant over those in the pills.
