Chinese forest frog, its Latin name is Rana temporaria chensinensis, they only grow in Changbai mountain area, the northeastern of China. The eggs of Rana temporaria chensinensis could be as a drug because it contains many special compositions related modulate incretion. But there is no reports about the anticancer protein from the eggs of Rana temporaria chensinensis in the world recently. In America , some researches showed that some proteins from frog eggs,an analog of ribonucleases, displayed anticancer properties recently. One of these frog proteins was currently as a cancer chemotherapeutic in clinical trials. Recent research about them are trying to show the anticancer mechanism of frog proteins, which may lead to a new class of drugs. S180 cells were inoculated hypodermically at the concentration of 1×106 cells per mouse and the protein solution of forest frog eggs was injected into the muscle of mice everyday. After continuous injection for 15 days, the inhibition rate of tumor was determined. The induction of tumor cell apoptosis of anticancer protein of eggs of Rana temporaria chensinensis could be detected by morphological observation, MTT colorimetric estimation, Flow cytometer detection. Anticancer protein from the eggs of forest frogs could significantly inhibit the growth of Hca-f cancer cells and S180 tumor cells, and could induce the apoptosis of K562 cells. We have determined the cell membrane was damaged, cell transmuted or produced gemmae, constricted region and apoptotic bodies, and the transparency and brightness of cells was declined after the K562 cancer cells were treated with eggs protein for 72 hours. The proliferation function of cancer cell and the number of active cells decreased. Cell crenation, condensation and margination of nuclear chromatin by cytoplasimic hypervacuolization could be seen by morphological observation. The results by flow cytometer showed that significant apoptotic peak. DNA content in S phase is less than in G1 phase,it showed that this anticancer protein from the eggs of forest frogs could block the checkpoint from G1to S phase in cell cycle.
