Human B Lymphocyte Stimulator(hBLyS) is a new member of tumor necrosis factor(TNF) superfamily. BLyS has three receptors BAFF-R, BCMA and TACI, all of them are expressed in B cells. It stimulates proliferation of, and immunoglobulin production by, B cells in the presence of Anti-IgM or Interleukin-4(IL-4), BLyS plays an important role in the regulation of humoral immunity. BLyS also plays a role in autoimmune diseases. Clinical studies suggest a potential role for BLyS in the progress of lymphoid malignancies.Chimeric toxin is a heterogeneous molecule that is composed of homing molecule and cytotoxic molecule. Homing molecules are commonly antibodies and cytokines, toxins are almost super-antibodies and inhibitors of protein synthesis. Chimeric toxin has many advantages in tumor therapy: selective killing tumor cells, high cytotoxity effect, no drug fastness.In this paper we describe in detail study on the heredity stability of a recombinant plasmid pET30a/hsBLyS in Ecoli BL21 and construction , expression and activity detection of the chimeric toxin hsBLyS-PE40.
