BACKGROUD & OBJECTIVE: The tiny RNAs are termed "shortinterfering RNAs"(siRNAs) depending on their origin. They down-regulategene expression effectively by binding to complementary messengerRNAs(mRNAs) and triggering mRNAs degradation (RNAi). The currentresearch is designed to investigate whether siRNAs can inhibit theexpression of Smad7 gene. METHODS: Gene transfection and Northern blot are performed todetermine the effect of gene inhibition of Smad7 by RNA interference inBEP2D and BERP35T2 cells. Meanwhile,fluoroscopy is used to evaluateimpact of GFP siRNAs on GFP expression. RESULTS: It could be concluded that RNAi can inhibit specificexpression of Smad7 gene and GFP gene. CONCLUSION: The results lay foundation for further study onfunction of Smad7 gene and TGF-β signal transduction pathway.
