| Nuodikang ( NDK ) is manufactured in Sichuan Rhodiola WeiguangPharmaceutical Co., Ltd. It is a new national Chinese traditional medicine, whichis made from rhodiola sacra s.h.fu in high plateau and has obvious curative effecton diseases of heart and blood vessel, but its mechanism is still not obvious. Toinvestigate the effects and mechanisms of this medicine on heart and blood vessel,this experimentation was carried through following three aspects.1. Protective effects of NDK on isolated Guinea pig heart with ischemia-reperfusion injuryObjective: Investigating protective effects and mechanisms of NDK onisolated Guinea pig heart with ischemia-reperfusion injury. Methods: Medicinehad been given to Guinea pigs through mouth then into stomach for 5 days(twiceone day), and then their hearts were isolated and perfused on a Langendorffapparatus with K-H liquid to be made a model of ischemia-reperfusion injury,recording changes of monophasic action potential(MAP),ECG and cardiaccontractility with biological signal analytical system. At the end of reperfusion, wealso recorded the swelling degree of hearts and mensurated the content ofglutathione peroxidase(GSH-PX),suveroxide dismutase (SOD),malondialdehvde(MDA) and lactate dehydrogenase(LDH). Results: (1)NDK could enhancedepolarization amplitude and velocity during 0 phase of MAP,prolong 2 phase butnot all the duration,decrease the drift of ST segment of ECG during the course oflow flow perfusion,increase cardiac contractility and reduce the incidence ofreperfusion arrhythmias。(2)NDK could decrease the swelling degree of hearts,enhance activities of GSH-PX and SOD and decrease the content of MDA andLDH。(3)All the effects were dose-dependent。Conclusions: NDK could enhanceNa+ inward flow in INa and Ca2+ inward flow in ICa-L, decrease oedema, decreaseperoxidation of lipid, reduce the incidence of reperfusion arrhythmias and so on, sothe ability of anti-reperfusion injury of myocardial cells was enhanced.2. Effects of NDK on hemodynamics in acute myocardial ischemia and heart failure ratsObjective:To investigate the effects of NDK on hemodynamics in acutemyocardial ischemia and heart failure rats and study its hemodynamic mechanisms.Methods: Medicine had been given to rats through mouth then into stomach for 5days(twice one day), and then acute myocardial ischemia was induced bypituitrin,and after 20 minutes acute heart failure was induced by pentobarbitalsodium. We recorded hemodynamic variables with biological signal analyticalsystem. Results:(1) Acute myocardial ischemia induced by pituitrin was opposedby administration of NDK: it decreased ST segment drift on ECG, increased HRand decreased LVEDP; LVSP,+dP/dtm and -dP/dtm of NDK group were morethan others,and the effects were dose-dependent. (2) Acute heart failure inducedby pentobarbital sodium was opposed by administration of NDK: it increasedblood pressure,HR,LVSP,+dP/dtm and -dP/dtm, and the effects weredose-dependent. Conclusions: NDK could enhance cardiac contractility and relaxblood vessel, so it could improve hemodynamics of acute myocardial ischemiainduced by pituitrin and heart failure induced by pentobarbital sodium in rats.3. Effects of NDK on isolated rat thoracic aortaObjective:To investigate the effects of NDK on isolated rat thoracic aortaand study its possible mechanisms. Methods:The isolated thoracic aortas thatwere removed from Wistar rats were cut into many rings and they were studied inisolated organ bath. Isometric tensions of the vascular rings were measured withbiological signal analytical system. Results:(1)NDK did directly cause relaxationresponses of vascular rings. (2)NDK significantly inhibited the contraction of thevascular rings induced by potassium chloride.(3)The vascular rings werepreincubated by NDK in K-H fluid without calcium but with 60mmo1/1 potassiumchloride, then the contraction of the vascular rings induced by cumulative calciumchloride was inhibited. Conclusions:NDK could inhibit the contraction of thevascular rings through adjusting calcium channels. |
