Chinese Medicine Wedelia Chemical Composition And Anti-viral Activity

Wedelia chinensis Merr. is Wedelia. plant of Compositae. It is used for the treatment of diphtheria, chincough, diarrhea, hemorrhoids, injuries from falls, faucitis, measles. Now it is preferably used for the treatment of hepatitis and stomatitis in clinic.However, few studies on the chemical constituents and bioactivities of this plant has been reported. In order to develop the natural resources, search for its active components, extend the clinical effects and make a new drug from Traditional Chinese Medicine, this dissertation dealed with the chemical constituents and bioactivities of Wedelia chinensis Merr.The pieces of Wedelia chinensis Merr. were extracted successively with 90% ethanol. The ethanol extract was dissolved in water, and re-extracted with petroleum ether, chloroform, ethyl acetate, n-butanol. Thirteen compounds were isolated from the chloroform and ethyl acetate section of ethanol extract of Wedelia Chinensis Merr.. On the basis of chemical and spectral analysis(UV, IR, MS, 1HNMR, 13CNMR, DEPT, HSQC, HMBC, 1H-1HCOSY), the structures of twelve compounds were identified as: Luteolin-6-C- β -D-digitoxoside(F6- II ), 7-Methoxy-2 -hydroxy-5,6-methylenedioxy isoflavone (L3-IV), Oleanolic acid-11,13(18)-dien-3-O-β -D-glucuronopyranoside(F7-III), leanolic acid-11,13(18)-dien-3-O-β -D-glucuronopyranosylmethylester(F7-I), 9(E),11 (Z),13(E)-Dien-8,15-dione-octadecoic acid(L3- V ), Chikusetsusaponin- IV a(Fll- I), Calenduloside E(F7- II ), Chrysophenol-8-O- β -D-glucopyranoside(F6- I ), Rhaponticin(F10-II), α -spinasterol (L2) , β-sitosterol (L3) , Lacceroic acid(Ll), Among them, F6-II, L3-IV, F7-I , F7-III, L3-Vare new compounds and F11-I , F7-II, F6-I , F10-II, L1, L2 are isolated from Wedelia. for the first time, and the identification of coumpound F6-III is now in press.In pharmacology, Anti-virus activity of three compounds have been found by preliminary anti-HIV, HIV-1 IN and HBV activity-evaluating. Compound F6-I and F7-III have significant anti-HIV activity, their values of EC50 are 25μg/ml and 50μg/ml. Compound F7-II and F7-III have anti-HIV-1 IN activity, their values of EC50 are 43.019μg/ml and 39.813μg/ml. Compound F7-III have anti-HBeAg activity, EC50 is 13.19μg/ml.
