| To utilize deeply and effectly the source of Platycodon grandiflorum (Jacq.)A.DC, based on the theories and methods of Physical chemistry and Natural medicinal chemistry and Pharmacology, using platycodins and prosapogenins as the material, we researched the relationship of surface activity between platycodins and prosapogenins, the relationship of hemolysis between platycodins and prosapogenins, the relationship between surface activity and hemolysis.Platycodins has extensive pharmacological actions,the surface activity is used for removing the phlegm and relieving cough. But platycodins is hemolysis toxic,the paper is dued to research the relationship between hemolysis and chemical structure,and provide the scientific foundation for reducing hemolysis activity of platycodins. We are dued to search for components which have pharmacological activities similar or superior to the corresponding natural products but less toxic.By Silica Gel Column,low pressure Rp-C18 column,Centrifugal thin layer Chromatography,high speed countercurrent chromatography(HSCCC), and TLC in three different developments, 5 compounds were isolated from prosapogenins, they are compound I~V; 6 compounds were isolated from platycodins, they are compound VI~XI.Eleven compounds were identified by TLC,IR,1H-NMR,13C-NMR,MS and Physicochemical evidence of compound I [3-O-β-D- glucopyranosylpolygalacic acid],compound II [3-O-β-glucopyranosylplatycodigenin],compound III [3-O-β- gentiobiosylplatycodigenin] , compound IV [methyl 2-o-methyl-3-O-β-D- glucopyranosyl platycogenate A],compound V [3-O-β-D-glucopyranosyl-(1→6)-β-D-glucopyranosyl- ( 1→6 ) -β-D-glucopyranosyl -2β, 3β, 16α, 23,24–pentahydroxyoxyolean -12-en-28-oic acid],compound VI [platycodin A],compound VII [platycodin C] , compound VIII [platycoside E] , compound IX [2"-O- acetylpolygalacin D2 or 3"-O-acetylpolygalacin D2],compound X [platycodin D3],compound XI [platycodin D ].Temperature 20℃, different concentration water solutions of platycodins and... |
