The incidence of deep-seated or disseminated fungal infections has increased steadily over the past two decades. Therefore the work of seeking for new antifungal drugs is becoming more and more urgent, which turns into a work with great realistic significance. β -1,3-glucan is the largest and most important component in the cell wall, which is responsible for 40-50% by weight of the cell wall. Change on the quantity and quality of them will exert tremendous influence over the cell itself. β -1,3-glucan synthase, synthesizing β -1,3-glucan, exists in cell membrane and is one of the most important enzymes for fungal cells. The cell wall of fungi is a unique component in lower eucaryote and mammals lack such component, consequently taking the β -1,3-glucan as the target to screening new antifungal drugs can avoid the disadvantages such as toxicity to human body , cross resistance and also has creativitiy.Extracting crude membrane protein from the Candida albicans ATCC10231 broth, measure its concentration, size and activity. Establishing the screening model of inhibitors against β-1,3-glucan synthase. Adopting this model we screened microbial fermentation broth of 3200 samples. We got 112 positive samples from primary screening, and through later reptitive screening 6 positive samples are found.
