| Objective: To study the effect of lentina(LNT) on the expression of vascular endothelial growth factor(VEGF) and microvascular density(MVD) in Lewis lung cancer-bearing Balb/C mice .Method: 40 Balb/C mice were devided into 4 groups randomly(n=10 each ), Lewis lung cancer cell were implanted into 4 groups by subcutaneous injection. After 48h, the 4 groups mice were given 0.9%NaCl 0. 2mL, DDP 0. 2mL(20mg/mL) , LNT 5mg/kg and LNT 5mg/kg+DDP 0.2ml (20mg/mL) respectively, once a day. After the drug was given continuously for 10 days, the mice were killed by decapitation. The mice were weighed before decapitation, then observe vascular by microscope and vascular endothelial by electron microscope and the expression of VEGF protein and MVD in tumor by immuneohistochemistry method. All data was treated with SPSS10. 0.Result: 1.The mice of NS had tarnished fur, sarall amout of food, spirit sagged, moved targily, the weight increase significancely was less than other groups, the general state of mice in LNT+DDP was best; 2.Under light microscope, the nascent capillary in group NS was more dense than group LNT, group DDP and group LNT+DDP; 3.Under electron microscope , the wall of the nascent capillary in the tumor was flimsy , It was the pseudopod of endodermis cell which contacted endodermis cells, the dense between endodermis cells was wider than normal, the basement membrane was thin and relax. 4. VEGF protein was mainly stained in cytoplasm, nuclear membrane with brown color, the â…§ factor was mainly stained in cytoplasm, nuclear membrane with brown color, too. The expression of VEGF and the MVD in group LNT, group DDP and group LNT+DDP was significantly decreased as compared with that in group NS .Conclusion: 1. LNT can raise the life quality of Lewis lung cancer mice, depress the side-effect of western medicine . 2. LNT can inhibit the formation of the new blood vessels in the tumor of Lewis lung cancer mice. 3. LNT can inhibit the expression of VEGF in the tumor of Lewis lung cancer. 4. LNT can decrease MVD in the tumor of Lewis lung cancer. 5. Through inhibiting the formation of the new blood vessels in the tumor of Lewis lung cancer mice, decreasing MVD in the tumor and inhibiting the expression of VEGF in the tumor, LNT can inhibit metastasis of tumor. 6. Through inhibiting the formation of the new blood vessels in the tumor of Lewis lung cancer mice, decreasing MVD in the tumor and inhibiting the expression of VEGF in the tumor , LNT can raise the life quartily and inhibit the formation of the new blood vessels in the tumor. 7. LNT have co-operation with cisplatin. |
