The Brucea Anticancer Effective Parts Of The Chemical Composition And Method For The Determination Of The Study

Brucea javanica(L.)Merr is a traditional Chinese medicine, which distributed in the Hainan,Guangdong,Guangxi and Yunnan of China. The Chinese General Catalog of Herbs ('Bencao Gangmu') recorded that B. javanica could be used for the treatment of dysentery ,tenesmus and hemorrhoids.The chief constituents are quassinoids, with have antitumor ,antimalarial, antiamoebic activity.By means of column chromatography, PTLC,HPLC ,12 compounds were isolated from the antitumor active fraction of ethanol extract of the fruit of Brucea javanica(L.)Merr. On the basis of physico-chemical characters and spectroscopic analysis, their structures were identified as: brusatol (1), bruceine D (2), bruceine E (3), bruceine H (4), bruceoside A (5), bruceoside B (6), yadanzioside A (7), yadanzioside B (8), yadanzioside C (9), yadanzioside G (10), β-sitosterol (11), daucosterol (12).The content of brusatol in the antitumor active fraction of ethanol extract of fruits of B. javanica was quantified by HPLC. And it leads to the possibility of the quality control.The cytotoxicity in vitro of some isolated compounds towards 3 human cancer cell lines was tested. Compounds1,2,3,5,6 and one extract showed inhibitory activity towards human cell line SF188, U87, and MCF-7.
