| Objective:Since the incidence of diabetes mellitus ( DM ) is graduallyrising,diabetic nephropathy (DN)has become the leading cause of end stage renal disease in China.The patients have to depend on hemodialysis,peritoneal dialysis or kidney transplantation to maintain lives,which bring economic burden and stress to family and society.How to prevent and treat DN has become the focus. Transforming growth factor- β1 (TGF-β1) ,which has attracted more and more attention in recent years,has comprehensive biological functions,including regulating cellular proliferation and differentiation's well as the forming of extracellular matrix (ECM) .Present study is to investigate the effects of fluvastatin and emodin on expression of TGF- β 1 on diabetic rats models.Methods: One hundred and twenty sprague-dawley rats were randomly dividedinto six groups: normal control group (N group) ,diabetic control group (D group ) ,DN+fluvastatin group ( F group ) ,DN+emodin group ( E group) ,DN+fluvastatin combined with emodin group (FE group) .The diabetic rats models were induced by the single injection of streptozotocin (STZ) via coccygeal vein.The levels of blood glucose, plasma cholesterol, serum BUN, serum creatinine and 24 hours proteinuria analysis were measured by biochemistry method.The mRNA and protein levels of TGF- β1 in renal cortex were measured by reverse transcription polymerase chain reaction(RT-PCR) and immunohistochemistry.Results: (1) The comparison in weight:The decreasing of weight wasobserved in the diabetic rats,compared to the normal control group since the second week,and increased food and water uptake,as well as urine were alsoobserved on diabetic rats,in accordance with the typical symptoms of DM.
