Odoratum Lowering Blood Sugar Saponins And Their Quality Standards And Pharmacological Studies

Rhizoma Polygonati odorati, the dry root of Polygonatum odoratum (Mill.) Druce is a Chinese traditional medicine with lowering blood glycose, lowering blood lipid, lowering blood pressure, anticancer activities. The chemical constituents (steroidal saponins, cardiac glycosides, polysaccharides, flavonoids, sterols, alkaloids, amino acids) that have been isolated from rhizome Polygonati odorati. In recent years, the bioactivity of alcohol extraction attracted the interesting of chemists. By now, there is not any systemic comprehensive report about the quality standard of total saponins from rhizoma Polygonati odorati (YZZG).In this thesis, the alcohol extraction of rhizoma Polygonati odorati was studies, and two compounds have been isolated from the alcohol extraction by repeatedly chromatography on DIAION HP20 resin column, flash chromatography, Sephadex LH-20, normal and reversed-phase silica gel columns. On the basis of the chemical evidence and spectra analysis datum (UV, ESI-MS, ¹H-NMR, ¹³C-NMR, DEPT), the structures of two compounds were elucidated as following:3-O-β-D-glucopyranosyl-(1→2)-[β-.D-xylopyranosyl-(1→3)]-β-Z)-glu -copyranosyl-(1→4)-galactopyranosyl-25(R,S)-spirost-5-en-3β-ol ( POD-I ),3-O-β-D-gluco-pyranosyl-(1→2)-[β-D-xylopyranosyl-(1→3)]-β-D-glucopyranosyl-(1→4)-galactopyranosyl-2 5(R)-spirost-5-en-3β,14α-diol (POD-II).According to the Technical Requirements of Quality Standard on VI New Drugs(naturaldrugs).We chose POD-II as standard substance and reviewed the effects of different chromogenic agents on determining YZZG with UV spectrophotometry, and established the optimal color method. In order to prepare YZZG from rhizoma Polygonati odorati safely, appropriately, and effectively, alcohol extracting was investigated according to YZZG's characteristic in Chapter 4. Taking into account that active chemicals' content in the alcohol extraction is determined by alcohol concentration, volume, extraction repeats and time, We designed and performed a orthogonal test L₉(3⁴) to optimize the extracting procedure of YZZG, and purified the alcohol extract with DIAION HP20 resin column, the diversion rates of YZZG were computed, further established the optimum extracting and separating procedure of total saponins from rhizoma Polygonati odorati.The range of total saponins in rhizoma polygonati odorati was 3.35-6.34 mg/g, and the diversion rates were 50%⁹0%. The methods established for extracting, separating and determining total saponins of rhizoma Polygonati odorati would lay a foundation for the quality control of rhizoma Polygonati odorati.
