| Diabetes Mellitus (DM) belongs to"xiaoke"according to the thory of traditional Chinese medicine(TCM).TCM has a long history of treating diabetes since Zhongjing Zhang creat the differential treatment on diabetes by using Baihu plus Ginseng decoction to treat the type of middle jiao dryness hot diabetes and using Shenqi pill to treat the type of lower jiao deficient cold diabetes in East han dynasty.Chinese prescription treat diabetes has the character of multi-link and multi-target,it can comprehensive treatment for diabetes and effectively prevention and cure the complication,as a result,the Chinese prescription become a research hotspot in recent years.But research on the pharmacology of traditional Chinese medicine prescription is lagging,the action mechanism can not be explained clearly,so it obstruct TCM treat DM generalization and development.In order to research and develop a chinese prescription which can both reduce the blood glucose effectively and explain the action mechanism clearly,we perform the following experiment:1.According to the TCM prescription compatibility theory and the experiment result which we had before,we use the chinese medicine D,T,R to compose a TCM prescription-DT1, then perform the pharmacodynamic research on DT1 through obseving the influence of blood glucose,mice weight,drinking amount and dietary amount on diabetic mice model which was induced by STZ.After curing two weeks, the diabetic mice blood glucose was reduced obviously,the weight gain,the amount of drinking and dietary lower,the typical symptoms of diabetes were improved.2.According to the experiment results and the literatures,we separate and distill two different chmeical components from D,T,R,get six chemical components:Dg,Dt,Tg,Tt,Rg,Rt,use the TLC to ensure they were separated.Then we use the same method as we did pharmacodynamic study on DT1 to perform the pharmacodynamic research on the six chemical components,and assays the hepatic glucogen content,C-peptide and glucagon level in serum,observe the influence of pancreas pathologic damage to investigate the therapeutic mechanism.After four weeks treatment,all the chemical components reduce the diabetic mouses'blood glucose, typical symptoms of diabetes were improved,Dt reduce the blood glucose level more than others and it's effection is the most steady one.Through the pharmacology research ,we get the result below:Dg,Dt can increase the hepatic glucogen content obviously;Tg,Tt can improve the pancreas pathologic damage,increase secretion of C-peptide,reduce secretion of glucagon;Rg,Rt can increase the hepatic glucogen content.DT1 can improve the pancreas pathologic damage, increase secretion of C-peptide,reduce secretion of glucagon,increase the hepatic glucogen content.The result shows,DT1 via improve the diabetic mouse pancreas pathologic damage,increase secretion of C-peptide,reduce the secretion of glucagon,increase the hepatic glucogen content to reduce the diabetic model mice blood glucose.All the six chemical components Dg,Dt,Tg,Tt,Rg,Rt has therapeutic action for diabetic model mice,but they have different therapeutic effect and different action mechanism..In conclusion ,we validate the DT1 therapeutic effect on STZ induing the diabetic mice model,and combination the effective chemical components pharmacodynamic and pharmcology study to evaluate these effective chemical components function in the DT1 syntheticaly,filter the material basis of DT1,investigate the action mechanism.All the result provide the scientific basis for clinical medication,and make foundation of further research and development... |
