The purpose of this research is to explore the best technical conditions and micro emulsion preparing technology in extraction and refine of Gardenia, and the relative bio-availability of Panax pseudo-ginseng, a kind of traditional Chinese medicine in different administration routes in Tongluojiunao micro emulsion preparation, a traditional Chinese medical formulae. In this experiment we found that:1 Research on percolation method, a kind of extraction technology on GardeniaAdopting the percolation method to extract, by carrying out the L9(34)cross experiment, taking the particle size of medical material (A), concentration (B) and dosage (C) of alcohol as the factors, moreover, yield of powered extract and content of jasminoidin as the indicators we screen the best technical conditions. The conclusion is: taking the content of jasminoidin as the indicator, the sequence of each factor is A>B>C, the analysis of variance shows that: all the three factors, A, B and C have the significance. Therefore, the best technical condition for percolation extraction of Gardenia is No. 24 screen mesh Gardenia powder with 8 times of 70% alcohol. The yield of jasminoidin is 4.099% and powdered extract is 20.36%.2 Research on Refine technology on GardeniaOptimization of alcohol extracting and water precipitating purifying technology of Gardenia: taking times of water added(A), content(B) and PH(C) as the factors, three levels for each, adopting the L9(34)Cross experiment, moreover, yield of powered extract and content of jasminoidin as the indicators we screen the best technical conditions for diluting by adding water. The conclusion is: taking the content of jasminoidin as the indicator, the sequence of each factor is B> C> A, the analysis of variance shows that: all the three factors, A, B and C do not have the significance. Taking the convenience of technology in actual production into consideration (4.6 is the original PH value of medicine liquid), therefore we confirmed that the medicine liquid condensed to relative density 1.10, 4 times of water to dilute and precipitate is the water precipitating refine technology for Gardenia. Extraction rate of effective ingredient is 89.97%.3 Research on optimization of activated carbon column elution requirement on Gardenia extractTaking concentration of medicine liquid(A), concentration of alcohol for elution(B), eluting reagent dosage (C)and eluting velocity (D) as the factors, three levels for each, L9(34)Cross experiment adopted, moreover, taking yield of powered extract and content of jasminoidin as the indicators we screen the best technical conditions for column chromatography by using activated carbon. The load of activated carbon column in this cross experiment is 50g. The conclusion is that: factor A and B have the significance while C and D have not. Therefore, the best technology of activated carbon elution is medicine liquid with relative density 1.10, 10 times of 70% alcohol and elution velocity 1ml/min. Extraction rate of effective ingredient is 77.19%.4 Research on preparation technology of Tongluojiunao micro emulsionResearch on empty emulsion prescription: taking tween-80 as the surface acting agent, 1, 2- trimethylene glycol as the assistant surface acting agent, isopropyl myristate the oil phase. Mixing tween-80 and trimethylene glycol by the volume ratio (Km) of 4:1, 3:1, 2:1, 1:1 and 1:2, and then mixing the mixed liquor with isopropyl myristate by quality ratio respectively of 12:1, 9:1, 8:2, 7:3, 6:4, 5:5, 4:6, 3:7, 2:8 and 1:9, stirred to well distributed, stirred on the magnetic stirring apparatus and titrated each mixture by deionized water while stirring to prepare the micro emulsion, the system changed from turbid to transparence shows micro emulsion formed. Drawing the pseudo-three element phase diagram. The micro emulsion ratio confirmed is tween-80: 1,2- trimethylene glycol: isopropyl myristate : deionized water=9: 3:1:30. Observed under the transmission electron microscope, inner phase is round and well distributed. The average particle size measured by laser particle size determination apparatus is 37.1 nm.5 Research on relative bioavailability of Panax pseudo-ginseng in different administration routeDetermination of Panax pseudo-ginseng blood concentration in rabbits in different administration route: taking panaxsaponin Rg1 as the indicator, micro emulsion and water p.o. by rabbits, compared with Xueshuantong injection by margin ear vein injection, the bioavailability of micro emulsion p.o. is 60.03%.
