Asparagus officinalis Linne, with the scientific name of Asparagus, adscripts to Liliaceae. It has plenty of flavonoids compounds named rutin. It is well known that the flavonoids is one of the major activating substances in medicinal plants, and have many functions of antiinflammatory, immunomodulatory, anticancer, antimicrobial, antiviral, antioxidant, and antithrombotic,et al. Therefore, the development and application of Asparagus officinalis Linne had already drawed a widely public concern in recent years.The RP-HPLC method is adopted to determining the rutin contents in different parts of Asparagus Officinalis Linne. The rutin contents were extracted by ultrasonic wave treatments before column (ZORBAX Eclipse XDB-C18 (4.6×250mm) ) derivatization, and then were detected with ultraviolet detection at 360nm using a mobile phase of methona-phosphoric acid solution. The result showed that the top part get the highest level of rutin content which reached to 1.762 mg/g, and then middle part reached 1.030 mg/g and finally came the base part 0.320 mg/g.Three extraxtion methods such as ultrasonic wave treatments,50% ethanol reflux with agitation and extraction with soxhlet extraction process were used to get the asparagus flavonoids of top part, during the extraxtion rutin was used as reference substance and the detecting wavelength is 510nm.The experiment result showed that the extraction efficiency were 42.956%,30.7752%,6.4376% respectively, and each the flavonoids contents were 0.949%,0.8383%,2.75%.The research of 400μg/mL, 200μg/mL, and 100μg/mL extractions'effects on LPS stimulated macrophage RAW264.7 NO release showed that extraction with Soxhlet extraction process in 400μg/mL could reduce LPS stimulated macrophage RAW264.7 NO release significantly (p<0.05); LPS stimulated macrophage RAW264.7 be treated with that extraction with Soxhlet extraction process in 400μg/mL,200μg/mL,100μg/mL .Griess method and Elisa method were used in the detection of NO release and content of IL-6, and the results declared that the extraction with Soxhlet extraction process in 400μg/mL could reduce NO release notability, and 400μg/mL,200μg/mL,100μg/mL could reduce contentof IL-6 notability in a dose-dependant manner.Using the model of DSS induced colitis established in 1990 by Cooper et.al, eight Blab/c rats (the number of males and females are the same) treated with 5% DSS seven days in comparison with those in the controls the DAI,activity of MPO,the length of colon were significantly changed. 32 Balb/c rats (the number of males and females are the same) were divided into 4 groups randomly which named A, B, C and D respectively. The mice group A was treated with distilled water, meanwhile the mice group B, C, and D were treated with 6mg/mL,10mg/mL,14mg/mL extraction of Soxhlet extraction process respectively. And then both were given 0.5ml per day until eleven days while 5% DSS treated seven days. The experiment results are given in the following: (1) the DAI value decreased obviously after 8 day, and after 11 days group D had the lowest DAI value; (2) the activity of MPO of group B, C and D were all reduced in comparison with group A, and group D was dropped obviously in comparison with group A; (3) the length of colon of group B, C and D were longer in comparison with group A, but not notability; (4) Extraction treated colonic tissue H.E stain showed that there were some ameliorations in a certain extent in the area of ulceration, soakage of inflammation cells, turbulence of epithelium and damage of the recess, and among them the amelioration of group D was the best.
