| Objective: First,to study the effect of Ginseng water-solubility total protein on African green monkey kidney cells (Vero), murine normal fibroblast cells (L929), human embryo lung diploid cells (2BS) and human throat cancer cell (Hep-2) in cell proliferation .Second, to study the effect of antler collagen protein(type I ) on Vero, L929, human cervix cancer(Hela), rat osteoblast cells and epidermic cells.Method: First ,we used such methods as distillment of neutral buffer and thiamine hierarchical-deposit to seperate ginseng water-solubility protein,which was then added to African green monkey kidney cells (Vero), murine normal fibroblast cells (L929), human embryo lung diploid cells (2BS) and human throat cancer cells (Hep-2). The cell growth effect was detected by MTT( Mono-nuclear cell direc cytotoxicity assay).Second ,we added Vero, L929, Hela, rat osteoblast cells and epidermic cells on antler collagen-protein-coated substrate boards and uncoated substrate ones. The cell growth effect was detected by MTT after a period of cultivation.Results: Ginseng water-solubility total protein obviously inhibits the cell growth of Vero and the Hep-2. At low concentration, ginseng water-solubility total protein promotes the cell growth of L929, especially at 1g/ml (P < 0.001). However,it inhibits the cell growth of L929 when the concentration reaches 500g/ml (P < 0.01). The total protein doesn't significantly affect the growth of 2BS cells. The antler collagen protein(type I ) greatly promotes the proliferation of Vero , L929 and Hela. Its proproliferation effect is stronger on rat osteoblast cells than on epidermic cells.Conclusion: Ginseng water-solubility protein exerts different influence on different cells. Our result indicates that although ginseng is a tonic, people cannot take it at will . Antler collagen protein can greatly help cells paste to the wall and improve proliferation, indicating that antler collagen protein is a good matrix material in terms of improving adhesion and cell growth. |
