Studies On Chemical Constituents Of Polygala Tenuifolia Willd

Radix Polygalae is one of common traditional Chinese medicine in ChinesePharmacopoeia volume i (2005 edition). The source is from the root of the perennial herbagepolygala tenuifolia Wild or polygala sibirica L family polygalaceae.The medicinal material ismainly produced in the areas of Shanxi,,Shanxi,Hebei,etc.It mainly used as an expectorant,tonic, sedative and dementia preventing agent. On the review of chemical constituents ofpolygala tenuifolia Wild. and its pharmacological activities,we investigated constituents oftriterpenoid saponin of Polygala tenuifolia W.Five compounds were isolated from it by usingopen-column(silica gel, macroreticular resin, Sephadex LH-20,ODS), preparative HPLCmethods et al. On the basis of spectroscopic analysis,including ¹H-NMR,¹³C-NMR,ESI-MS,HMBC,HMQC,¹H-¹H COSY,TOCSY and chemical methods, the structure of fourcompounds were identified as 3- O -β-D- glucopyranosyl -presenegenin-28- O -β-D-xylopyranosyl (1→3)β-D-xylopyranosyl- (1→4) [β-D-apiofuranosyl (1→3)]α-L-rhamnopyranosyl (1→2) [α-L-rhamnopyranosyl (1→3) ] (4-O-p-methoxycinnamoyl)-β-D-fucopyranosyl ester (â… );3- O-β-D-glucopyranosyl-presenegenin -28- O -α-L-rhamnopyranosyl (1→4) [β-D-apiofuranosyl (1→3)]-α-L -rhamnopyranosyl (1→2) -β-D-fucopyranosyl ester (â…¡);presenegenin-(3)-O-β-D-glucoside(â…¢); 3-O-β-D-glucopyranosylpresenegenin28-(2)-O-β-D-xylopyran osyl(1→4)-α-L-rhamnopyranosyl (1→2)-β-D-fucopyranosyl ester (â…£) (Polygalasaponinâ…©â…©â…§); compoundsâ… andâ…¡are new compounds.
