PART-ⅠThe expression of TREM-1 in the patients with endometriosisAIM:TREM-1(triggering receptor expressed on myeloid cells)-1 is a novel activating receptors of the Ig superfamily. In this study, we aimed to evaluate the levels of TREM-1 in peripheral leukocytes, soluble TREM-1(sTREM-1) in peritoneal fluid (PF) and serum, as well as the role of TREM-1 on the pathogenesis of endometriosis (EMS).METHODS:The level of sTREM-1 were measured by ELISA in women with or without EMS, and the expression of TREM-1 mRNA in peripheral leucocytes from sixwomen with EMS and sixwomen without EMS were detected by using real-time RT-PCR. And then, using immunohistochemical analysis, we evaluate the expression of TREM-1 in the ectopic endometrium of women with endometriosis.RESULTS:There were significant differences in sTREM-1 levels in peritoneal fluid and serum between EMS patients and control (p<0.01). The expressions of TREM-1 mRNA in the women with and without EMS were 0.765±0.022 and 0.695±0.035, respectively. There was also significant difference between the two groups (p<0.05). Furthermore, there was noTREM-1 expression in the ectopic endometrium.CONCLUSION:The expression of TREM-1 in peripheral leukocytes and sTREM-1 in PF and serumof those women with EMS were up-regulated, suggesting that TREM-1 might play an important role in the occurrence and development of EMS.PARTⅡThe effects of TNF-αand sTNFRⅠon stormal cells of endometriosisAIM:To explore the effects of TNF-αand sTNFRⅠon cultured eutopic endometrial stroma cells from endometriosis patients and find new biological therapy to endometriosis.METHODS:Stroma cells from eutopic endometrium were cultured in vitro. TNF-α(0, 0.1, 1, 10, and 100 ng/ml) was added to the medium in dose-responds experiment. TNF-alpha (1ng/ml) was added for 0, 4, 8, 12, 24, 48 and 72 hours in time-course experiment. TNF-α(0.1, 1 and 10 ng/ml) and sTNFRⅠ(2μg/ml) were added for 12 hours. Then the supernatant were collected to evaluate the productions of IL-6 and MMP-3.RESULTS: The productions of IL-6 and MMP-3 in the supernatant of stroma cells from endometriosis were promoted by TNF-alpha in a dose-dependent and time-dependent manner. The productions of IL-6 and MMP-3 in the supernatant of stroma cells from endometriosis were promoted by TNF-alpha were decreased by sTNFRⅠ.CONCLUSION:TNF-αmay promote the attachment and invade of stroma cells from women with endometriosis though inducing the expression of IL-6 and MMP-3 and play a central role in the pathogenesis of endometriosis. sTNFRⅠmay be the potential target for biological therapy to endometriosis.
