In clinic, the vigour-flushing and blood-activating prescriptions were widely used to treat diseases correlation with immunization disorder, such as that of the liver, the sugar diabetes, the apoplexy, the coronary heart disease, the gynecologist, the aged diseases et al. So the purpose of this research is to study the immune regulation mechanism of vigour-flushing and blood-activating prescriptions with modern immune technology and molecular biology knowledge. This research can rereal the pharmacology of this prescription and provide theoretical base for reasonable medication in clinical. By the syudy, new thoughts and methods for the research on the essential of vigour-flushing and blood-activating prescriptions can be provided.ObjectiveTo explore the aetiological factor on treating aged diseases in theory. And to observe the role of immune gaidelines about vigour-flushing and blood-activating prescription acting on the rats made decrepitude by D-galactose, Try to explain the action mechanism of vigour-flushing and blood-activating prescription.MethodsThe sixty healthy male mice were separated into six groups, they were normal groups, decrepitude model groups, Vitamin E groups, and the else three different dosages vigour-flushing and blood-activating prescriptions (2g/ml, 1g/ml, 0.5g/ml) groups, the three different dosages Chinese traditional medicine groups and Vitamin E groups would be dosed after they had been made models ten days.Results1. The effect on the weight index and ultrastructure of the thymus.Obviously shrink and regression in the thymus appear in the decrepitude model groups. The structure was blur and the cells in immune organ decreased and diffluenced, chromat becomed fade. Given different dosages vigour-flushing and blood-activating prescriptions differently lessen the variety of these mice, and the weight index was improved with dose increase, the effect of the highest dosage prescriptions was better than vitamin E groups.2. The effect on the SOD activities in the thymus and spleen of mice.The SOD activities in the thymus of mice were improved with close increase. Compared with model groups, the different dosages prescriptions, normal groups and vitamin E groups had distinct difference (p<0.01),compared with vitamin E groups, the different dosages prescriptions had no difference(p>0.05), and there was no difference among the different dosages prescriptions.The SOD activities in the spleen of mice were improved with close increase. Compared with model groups, the different dosages prescriptions, normal groups and vitamin E groups had distinct difference(p<0.01); compared with vitamin E groups, the highest dosage prescriptions had distinct difference(p<0.05).3. The effect on the MDA contents in the thymus and spleen of mice.The MDA contents in the thymus and spleen of mice were depressed with close in crease. Compared with model groups, the different dosages prescriptions groups,normal groups and vitamin E groups had distinct difference(p<0.01), compared with vitamin E groups, the highest dosages prescriptions had distinct difference(p<0.05), there were distinct difference among the different dosages prescriptions(p<0.01).5. The effect on the rate of CD137 positive cells in the spleen cells suspension.The rate of CD137 positive cells in the spleen cells suspension was improved with dose increase. Compared with model groups, the different dosages prescriptions groups,normal groups and vitamin E groups had distinct difference (p<0.01 or p<0.05); compared with vitamin E groups, the highest dosages prescriptions had distinct difference(p<0.05), there was no difference among the different dosages prescriptions.Conclusion1. The decrepitude models was very successful. From the result of experiment, we can find: Obviously difference in the thymus index, the concentration of SOD and MDA in the thymus and spleen, the concentration of IL-2 in the spleen cell culture supernant, the rate of CD137 positive cells in the subtype of T lymphocyte cell, and the structure of thymus among the decrepitude model groups with the else five groups.2. The three different concentration vigour-flushing and blood-activating prescriptions groups can actively affect the thymus index, the concentration of SOD and MDA in the thymus and spleen, the concentration of IL-2 in the spleen cell culture supernant, the rate of CD137 positive cells in the subtype of T lymphocyte cell and the structure of thymus, so it can improve immune function of the aged mice. |
