| ObjectiveParkinson's disease, a parapymmid progressive neuro- degenerative diseases in centre nerve system, always invades middle-ages and older people and cause their disables. PD's mainly pathology change maintains the death of most of the dopamine neurons which project from tight zone of substantial nigra of midbrain to striatum. PD's mainly symptoms include stilly shacking, bradykinesia, rigidity muscles and abnormal posture and pace. Nowadays, the primary western medicines are L-dopa, dopamine agonists (DAs), monoamine oxidase inhibitors-B (MAOI-B), but these medicines could not prevent the progress of PD well and bring about a lot of side effects at the meantime. Operation and gene therapy could not treat the PD well, either. At present, acupuncture as a peculiar, safety, and convenient way has been applied in the treatment of PD and has good effection. The text will study the therapeutical effect and mechanism of the electroacupuncture on PD, which will offer experimental basis for applying electroacupuncture in clinic praction of the acupuncture moxibustion.MethodsPD model was established by microinjecting 6-hydroxy-dopamine into mesencephalic substantia nigra in the right brain. The same amount of normal saline was injected into the rats of control group. After 4 weeks, Apomorphine was injected into the rats abdominal cavity to induce rotation. The test were repeated every 7 days. Rats who had constant left rotation with a speed faster than 7r/min were selected as successful PD models. Successful PD model rats were selected for treatment and randomly divided into two groups, model group and electroacupuncture group. hcupoints of Fengfu and Taichong in EA group were punctured once a day for 14 days.1. Observation on behaviour: 0.5 mg/kg of apomorphine was injected into abdominal cavity to induce left rotation. The number of circles rotated within 30 mins after injection of apomorphine was recorded, twice a week for 2 weeks.2. Mitochondrial transmembrane potential, MMP. Flow cytometry measurement of rhodamine fluorescence intensity and fluorescence intensity values reflect the level of nerve cell mitochondria transmembrane potential sums.3. Neural cell adhesionmolecule, NCAM. The expressions of NChM and NCAM mRNA were determined in situ hybridization.4. Observe the changes of substantia nigra neurons ultrastructural by Electron microscope.All data were expressed through SPSS11.0 software system.Result1. Normal group and sham-operated group has no significant differences in MMP; Model group is minimum; EA group was slightly lower than normal group, but higher than the model group.2. In damaged side, the MMP of PD rats is significantly lower than normal group, sham-operated group and contra lateral side (P<0.01). In damaged side, The MMP of model group is significantly lower than EA group (P<0.01). The MMP of normal sides has no significantly difference (P>0.05).3. Denaturation of substantia nigra neurons in EA group is lower than model group.Conclusion1. EA can enable nerve cells to enhance mitochondrial membrane potential, delayed neuronal apoptosis, acupuncture neuronal cells play a protective role.2. EA can promote rat model of PD SNc neural cell adhesion molecule and its mRNA expression, thereby promoting rat model of PD synaptic plasticity play, and indirectly promote nerve cell regeneration.3.
