Human Breast Tumor Stem Cells Isolation And Preliminary Study On Biological Characteristics

Objective : Primary breast tumor cells which expressed CD44~+CD24-/LOW were isolated . To explore the ratio of CD44~+CD24-/LOWcells,CD44~+CD24~+cells and CD44-cellsMethods:Isolation breast tumor cells from breast cancer tissue through mechanical dissociation and enzyme digestion.After cultivation of these cells CD44~+CD24-/LOWcells were isolated through immunomagnetic beads.Calculate the number of CD44~+CD24-/LOWcells,CD44~+CD24~+cells and CD44-cells by cell counting.Results:There had CD44~+CD24-/LOWcells in primary breast cancer cells . The ratio of CD44~+CD24-/LOWcells,CD44~+CD24~+cells and CD44-cells were 2.5%,30%,67.5% respectively. Conclusion:There had CD44~+CD24-/LOWcells in primary breast cancer cells.And the quantity of CD44~+CD24-/LOWcells were very low. Objective : Compare the reproductive activity and morphous characteristic of CD44~+CD24-/LOWcells,CD44~+CD24~+cells and CD44-cells. And to explore the difference of expressing of ER,PR and HER-2.Methods:Cultivate CD44~+CD24-/LOWcells,CD44~+CD24~+cells and CD44-cells.Draw growth curve of these cells.Using light microscope and electron microscope observation morphous characteristic. Immunohistochemistry determine the express ER,PR and HER-2 of CD44~+CD24-/LOWcells,CD44~+CD24~+cells and CD44-cells.Results : In the same condition, reproductive activity of CD44~+CD24-/LOWcells was the strongest one ,CD44-cells was the weakest one . There was no difference of morphous characteristic of CD44~+CD24~+cells and CD44-cells.CD44~+CD24-/LOWcells was obviously immature and CD44~+CD24~+cells and CD44-cells was mature CD44~+CD24-/LOW cells did not express ER,PR and HER-2 but tumor tissue express ER and did not express PR,HER-2. Conclusion : CD44~+CD24-/LOW cells was the source tumorous growth . CD44~+CD24-/LOWcells was obviously immature ,CD44~+CD24-/LOWcells maybe the tumor stem cell.
