Objective: An acute spinal cord injury (SCI) model of rat was made bymodified Allen's weight dropping technique. The objective of this studyis to study the protective effects of breviscapine on the acute spinalcord injury.Materials and methods:52 SD rats were randomly divided into threegroups, that was, the injury group (n=24), the therapy group of bre-viscapine(n=24)and the sham-operation group(n=4).The SCI animal modelwas injured by using a modification of the Allen's weight droppingtechnique. Animals were anesthetized with 3% pentobarbital (30mg/kg) byintraperiton- eal injection. After sterilization of the injury group andthe therapy group, T6-8 laminectomy was performed. Then the dura materof spinal cord was exposed. A metal stick weighed 10g was dropped freelyperpendicularly from 5cm height through a hollow steel pipe to strike theT7 spinal cord producing a model of acute SCI. The shamoperation groupomitted the weight dropping procedure. After the surgery, the therapygroup was given breviscapine 5mg/kg.d through intraperitoneal inje-ction. The sham-operation group and the injury group were given the sameamount of saline instead. Immunofluorescences method was applied todetect the protein expression of NF-κB P65,MCP-1,Bcl-2; TUNEL methodwas used to detect the cell apoptosis; Immunohistochemistry method wasapplied to detect the protein expression of PARP-1.Results: In breviscapine group, the expressin of NF-κB P65,MCP-1,PARP-1 protein was lower (P<0.05), while that of Bcl-2 protein expressionwas higher than in the control group(P<0.05). The number of apoptoticnerve cells was significantly less in breviscapine group than that in the control group by TUNEL(P<0.05)and the degeneration of neurons andneurofibril was milder than in the control group.Conlusion Breviscapine could protect neurons and decrease theapoptosis of neurons.
