Objective:To investigate the effect of PNS from the point of view of experimental on the bone marrow stem cell transplantation ischemic hind limb ischemia in rat model of angiogenesis site, and through ischemic hind legs of rats Doppler blood flow and the content of serum VEGF to explore its role of the molecular mechanism .Methods: The rats were randomly divided into four groups, namely the normal group, ischemia group, stem cell transplantation, stem-cell transplantation + PNS group. Which ischemia group at different times by ischemia divided into five groups, namely ischemia for 4 hours, 3 days, 7 days, 14 days, 28 days group. Stem cell transplantation group was divided into 14 days of transplantation group and 28 days of transplantation group. Stem cell transplantation + PNS group was divided into 14 days drug treatment group and 28 days group. In addition to the normal group, the other groups were cut the femoral artery ligation of the left hind limb, built rat hind limb ischemia model.Transplanted stem cells for the stem-cell transplantation groups and Stem-cell transplantation + PNS groups. Hind limb ischemia intramuscular injection of stem cell separation. At the same time the groups began treatment.Give the stem cell transplantation groups PNS for intraperitoneal injection. Stem cell transplantation groups were injected with normal saline the same proportion. In use 14 days, 28 days, using laser Doppler blood flow measurement instrument detection of ischemic hind limb perfusion. In rat aortic blood, use ELISA to determination the changes in the serum of VEGF .Results: SPSS13.0 was used by the data processing, statistical methods using analysis of variance. In ischemic four hours of serum VEGF levels (15.83 +2.173 pg/ml) that is its normal value (30.79 +4.888 pg / ml) are significantly different (P <0.01). Of local perfusion (66.91 +1.909 pu) also dropped markedly, and continue to reduce ischemic first seven days. Ischemic 7-28 days, with the VEGF levels gradually rise, perfusion was also significantly increased, but still lower than the normal level (P <0.01). Stem cell transplantation serum VEGF levels and local perfusion both significantly higher than that of ischemia group (P <0.01). The serum VEGF levels and local perfusion of the stem cell transplantation + PNS group was significantly higher than that in both ischemia group (P <0.01) and stem cell transplantation group (P <0.01). The local perfusion of the stem cell transplantation + PNS 28-days group (77.75 +3.009 pu) was even significantly higher than that of the normal group (P <0.01).Conclusions: Serum VEGF levels are sensitive indicators of ischemia, ischemic early (4 hours to 7 days) decreased to ischemic period (14 days to 28 days) increased slowly. Transplantation of bone marrow stem cells can improve the hind limb ischemia in the rat serum VEGF levels, and promote ischemic muscle tissue of angiogenesis. March 7 total saponin significantly increased serum VEGF levels, and increase the ischemic tissue perfusion, and this may be the promotion of bone marrow stem cell transplantation for the treatment of arteriosclerosis obliterans leg one of the mechanisms.
