Ganoderma lucidu, Poria cocos (Schw.) Wolf and Polyporus Umbellatus were chosen as subjects in this work. The optimal conditions of HPCE for detecting glucose were borax running buffer (80mmol/L), pH=10.2, separation voltage was 18 kV, injection time was 25s, detected wave was set at 254nm. The average contents of glucose in Ganoderma lucidum, Poria cocos (Schw.) Wolf and Polyporus Umbellatus were 3.817 7mg/g,15.378 5mg/g and 7.478 6 mg/g, respectively. The results show that HPCE is a fast and effective method which can be used to improve the quality specification for Ganoderma lucidum, Poria cocos (Schw.) Wolf and Polyporus Umbellatus.In this paper, the optimum extracting conditions of polysaccharide were also investigated by single factor test and the orthogonal experimental design. Compared with the traditional extraction methods, PSE was faster and more effective. In the optimal extraction process, the average contents of three kinds of medicinal fungus polysaccharides were 10.502 7mg/g, 8.696 5 mg/g and 7.868 4 mg/g, separately. In order to investigate monosaccharide composition and the molar ratio of Ganoderma lucidum polysaccharides, pachyman and polyporus umbellatus polysaccharides, three kinds of medicinal fungus polysaccharides were hydrolyzed. The monosaccharide composition of polysaccharide was analyzed by HPCE. The results showed that Ganoderma lucidum polysaccharides were composed by glucose, mannose and galactose, and the ratio of them was Glu∶Man∶Gal=2.806 3∶1∶2.191 0. While Polyporus umbellatus polysaccharides were composed by xylose, glucose, mannose, galactose and two kinds of unknown sugar. The ratio of the already known monosaccharide was xyl∶Glu∶Man∶Gal=0.464 2∶6.300 6∶1∶3.255 6. In addition, pachyman was composed by glucose, mannose galactose and another unknown sugar. The ratio of the already known monosaccharide was Glu∶Man∶Gal=2.656 5∶1∶2.361 7. The results show that analysis of monosaccharide composition of three kinds of medicinal fungus polysaccharides can be applied for the structural identification and content determination.
