Objective: To find the activitical parts and constituents, we had studied on the chemical constituents of leaves of Acer Saccharum and pharmacological experiment of leaves of Acer Saccharum, which will afford the scientific foundation and substantial basic of developing the new duge for antioxygenation and antibacterial activity.Methods: The constituents were extracted by leaves of Acer Saccharum with ethanol, and extracted with ethyl acetate and n-butanol, then isolated and purified by silica gel column chromatography and recrystalization, some structures were identified through spectroscopic analysis (1H-NMR,13C-NMR) and chemical methods. Samples of ethanol extracted were studied on pharmacological activities through measuring restraining ratio to DPPH, measuring SOD activities with Assay Kit-WST and measuring the reduction to NO with Griess for antioxygenation outside; through measuring activity of SOD and the quantity of MDA for antioxygenation inside body ;through measuring mice`ear tumidness for antibacterial activity inside. Determined flavones`and saponins` contents of leaves of Acer Saccharum by ultraviolet and visible spectroscopy.Results : Seven compounds were isolated from the plant, and three compounds structures were identified.Seven compounds were all isolated from Leaves of Acer Saccharum first.The compounds identified are 3-keton-ursane, 3β-hydroxy-12-oleanolic and 5-ene-7-ol -steroidnol. In experiment of Acer Saccharum (Leaves)5antioxygenation outside body, the restrain ratios to DPPH is 12.2±6.74%, activity of SOD is 0.27±0.003U/mg,the ratio of reduction to NO is 53.0±2.1%;in experiment of antioxygenation inside body ,the activities of SOD in blank group,model group, low-dose group and high-dose group are 16.20±3.00 U/ml,9.72±0.69 U/ml,39.70±8.57 U/ml,22.56±3.08 U/ml, through analysis of variance,there are highly marked differences of activities of SOD between low-dose group and high-dose group(P<0.01), there are highly marked differences of activities of SOD between high-dose group and model group(P<0.01), there are highly marked differences of activities of SOD between blank group and model group(P<0.01); the quantity of MDA in blank group,model group, low-dose group and high-dose group are 27.43±2.28 nmol/ml,35.96±5.08 nmol/ml,15.29±4.02 nmol/ml,17.70±8.14, through analysis of variance,there are marked differences of quantity of MDA between low-dose group and high-dose group(P<0.05), there are marked differences of quantity of MDA between high-dose group and model group(P<0.05), there are marked differences of quantity of MDA between blank group and model group(P<0.05). In antibacterial experiment,the mice`ear tumidness ratios of model group,high-dose group,low-dose group and control group are 114.4%,33.7%,20.4%,43.7%,through analysis of variance,there are highly marked differences of mice 5ear tumidness ratio between low-dose group and high-dose group(P<0.01), there are highly marked differences of mice`ear tumidness ratio between low-dose group and model group(P<0.01),there are highly marked differences of mice 5ear tumidness ratio between low-dose group and comparison group(P<0.01).The contents of flavones and saponins are 1.035mg/g, 1.105 mg/g in Leaves of Acer Saccharum.Conclusion : The componds which had been identified are tripenoids and steroidnol, and were isolated from the plant for the first time. Leaves of Acer Saccharum have high antioxygenation and antibacterial activity,high-doge group have better antibacterial activity than low-doge group, low-doge group have better antioxygenational activity than high-doge group.There are richer contents of flavones and saponins in leaves of Acer Saccharum.
