| Objective:To study the roles of central level of serotonin(5-HT) and serotonin 1A receptor(5-HT1AR) by electro-acupuncture.So as to provide study direction of exploring the central 5-HT1AR mechanism of anti-anxiety effect after morphine withdrawal by electro-acupuncture.So provides experimental foundation to resist relapse after drug addition withdrawal by acupuncture on clinic.Methods:Establishd morphine withdrawal model first,then randomly individed morphine withdrawal mice group into model group and electro-acupuncture group.The electro-acupuncture group mice were treated with acupuncture on acupoints "Baihui(DU20) and Sanyinjiao(SP6)".The treatment was once daily and 15 minutes everytime and lasted for 3 days.The behavioral tests were measured by the elevated plus-maze test(EPM),immunohistochemistry with image analysis technique were used to investigate the effect of electro-acupuncture on the change of the optical density(OD) value and grey value of raphe nucleus,hippocampi,amygdale and prefrontal cortex's 5-HT expressions and 5-HT1AR level in anxiety model after morphine withdrawal.Results:1.Comparing with control group(0.40±0.06,OT%;0.40±0.05, OE%),in the behavioral tests,the electro-acupuncture group's open-arms time (0.36±0.07,OT%)and the open-arms entries(0.37±0.05,OE%)were closely similar after been treated(P<0.01).The model group's OT%(0.22±0.05) and the OE% (0.24±0.03) were decreased obviously(P<0.01).2.The OD value of raphe nucleus and prefrontal cortex neurons's 5-HT in the electro-acupuncture group were increased significantly and gray value were obviously reduced,compared with model group (P<0.01,or P<0.05).Compared with control group,the electro-acupuncture group's 5-HT OD value and grey value did not have obvious varieties(P>0.05). However,the OD value and gray value of amygdale and hippocampi did not have significantly varieties in all groups(P>0.05).3.The OD value of amygdale,raphe nucleus and prefrontal cortex neurons's 5-HT1AR in the electro-acupuncture group were increased significantly and gray value were obvious reduced,compared with model group(P<0.01,or P<0.05).Compared with control group,the electro-acupuncture group's 5-HT1AR OD value and grey value did not have obvious varieties(P>0.05).Moreover,the OD value and gray value of hippocampi did not have significantly varieties in all groups(P>0.05).Conclusion:1.Anxiety effect after morphine withdrawal could be resisted by electro-acupuncture.2.The effect of resisting anxiety after morphine withdrawal by electro-acupuncture may relate to restrain releasing 5-HT of raphe nucleus's 5-HT neurons.3.The effect of resisting anxiety after morphine withdrawal by electro-acupuncture may relevant to enhance the level of 5-HT1AR of raphe nucleus,amygdale and prefrontal cortex neurons's 5-HT1AR. |
